鹅源性沙门氏菌分离鉴定及中药抑菌效果分析 被引量:2

摘要 鹅源性沙门氏菌病是严重危害鹅养殖业的一类细菌性疾病,同时也是一种潜在威胁人类食品安全的疾病。据报道,病鹅和病愈后的排菌鹅是该病的主要传染源,这些鹅会长时间携带病菌,并会在饲养管理不当的条件下大量排出病菌。其中以幼鹅患此病居多,发病幼鹅出现食欲差,精神萎靡,关节肿胀,跛行甚至导致死亡,多以与大肠杆菌混合感染为主。目前针对幼鹅沙门氏菌感染的病例多采用抗生素治疗,但药物残留现象也越来越严重。随着中医的不断强大国际认可度也在不断提升,中药在治疗细菌性疾病的应用上越来越广泛。采用不断纯化的方式用特异性沙门培养基分离幼鹅新鲜粪便中的沙门氏菌,利用琼脂斜面进行进一步纯化培养,最终勾取优势菌株通过PCR技术确定菌株类型。试用酒浸后超声波震碎并浓缩的中药采用传统的二倍稀释法测其最小抑菌浓度以期得到抑菌效果较好的中药。试验结果显示所选取的中药普遍对所取样品有抑菌效果五倍子、诃子、乌梅、防风、辛夷几味中药甚至具有高度敏感性。因中药讲究复方,各种中药炮制方法也不尽相同,故经其他的提取方式来抑菌其效果是值得进一步讨论的。可为以后中药在预防治疗鹅源性沙门氏菌病上提供理论依据。 Salmonellosis of goose origin was a kind of bacterial disease which seriously endangered the goose breeding industry and a potential threat to human food safety.According to the report,sick geese and cured geese were the main source of infection which would carry the bacteria for a long time,and in a large number of breeding and management of the disease under the conditions of discharge.Most of the young geese suffered from it.The young geese had poor appetite,listlessness,swollen joints,lameness and even death.Most of them co-existed with escherichia coli.At present,most cases of salmonella infection in goose were treated with antibiotics,but drug residues were becoming more and more serious.With the continuous development of Chinese medicine,the international recognition of Chinese medicine was also improving,and the application of Chinese medicine in the treatment of bacterial diseases was more and more extensive.In this experiment,salmonella in fresh goose feces was cultured with specific samana medium in the way of continuous purification.AGAR slant was used for purification and culture,and the strain was finally determined by PCR technology.The traditional two-fold dilution method was used to determine the minimum bacteriostatic concentration of the traditional Chinese medicine which was crushed and concentrated by μ Ltrasonic after soaking in wine in order to obtain a better bacteriostatic effect.The results showed that the Chinese medicine selected in this experiment generally had antibacterial effects on the samples,including Chinese gallnut,chebula,mume,fangfeng,magnolia,and even had a high sensitivity.Because the traditional Chinese medicine paid attention to the compound,each kind of traditional Chinese medicine processing method was not identical,therefore through other extraction method to suppress the bacterium deservs further discussion.This experiment could provide a theoretical basis for the prevention and treatment of salmonellosis of goose.
