清热凉血方对咪喹莫特诱导的银屑病样小鼠模型JAKs/STATs通路表达影响的研究 被引量:8

摘要 目的观察清热凉血方对咪喹莫特诱导的银屑病样小鼠模型皮损中酪氨酸激酶/信号转导因子和转录激活因子(JAKs/STATs)信号通路表达的影响。方法BALB/c雄性小鼠48只,随机分为空白组、模型组、雷公藤多苷组及清热凉血方低、中、高浓度组,每组8只,采用外用咪喹莫特乳膏诱导产生银屑病样皮损。肉眼观察小鼠背部皮损严重程度,光镜下观察皮损组织病理学变化;采用实时荧光定量聚合酶链式反应(PCR)法检测皮损组织中JAKs/STATs通路蛋白细胞因子信号转导抑制因子(SOCS1)、信号传导与转导激活蛋白3(STAT3)及相关细胞因子[白细胞介素(IL)-1、IL-6、干扰素(IFN)-γ、肿瘤坏死因子(TNF)-α)]mRNA表达水平。结果①肉眼观察发现清热凉血方低、中、高浓度组皮损均较模型组明显减轻,以高浓度组皮损好转最为明显,并与雷公藤多苷组相似;HE染色观察发现清热凉血方各组皮损表皮增厚较模型组轻,细胞浸润少,与肉眼观察结果一致;③清热凉血方各组SOCS1、STAT3、TNF-α、IL-6、IL-1、IFN-γmRNA表达较模型组降低(P<0.05),以清热凉血方高浓度组下降最为明显。结论清热凉血方可能通过抑制小鼠皮肤组织中JAKs/STATs信号通路的激活,减少TNF-α、IFN-γ、IL-1、IL-6的分泌,从而发挥治疗银屑病的作用,并且与低剂量组相比,中、高剂量组的这种作用更为明显。 Objective In order to observe the effects of clearing heat and cooling blood recipe on the expression of JAKs/STATs signaling pathway in the psoriatic mice models induced by imiquimot-induced skin lesions.Methods Forty-eight BALB/C male mice were randomly divided into blank group,model group,tripterygium glycosides group and low,medium and high concentration groups of clearing heat and cooling blood recipe,including eight mice in each group.External imiquimod cream was used to induce psoriasis-like skin lesions.The severity of skin lesions on the back of mice was observed by naked eye,and the histopathological changes were observed under light microscope.The mRNA expression levels of JAKs/STATs pathway proteins SOCS1,STAT3 and related cytokines[interleukin(IL)-1,IL-6,interferon(IFN)-γ,tumor necrosis factor(TNF)-α]in skin lesions were measured by real-time fluorescence quantitative polymerase chain reaction(PCR).Results①Visual observation showed that skin lesions of the low,medium and high concentration groups were significantly reduced compared with the model group,and the skin lesions of the high concentration group were the most obvious improvement,which was similar to the tripterygium wilfordii polyglycoside group;②HE staining observation found that the epidermal thickness of the skin of the clearing heat and cooling blood recipe group was lighter than that of the model group,with less cell infiltration,which was consistent with the visual observation results.③The mRNA expressions of SOCS1,STAT3,TNF-α,IL-6,IL-1 and IFN-γin each group were lower than those in the model group(P<0.05),and the most significant decrease was found in the high concentration group of the clearing heat and cooling blood recipe.Conclusion The clearing heat and cooling blood recipe may play a role in the treatment of psoriasis by inhibiting the activation of JAKs/STATs signaling pathway in mouse skin tissue,reducing the secretion of TNF-α,IFN-γ,IL-1 and IL-6,and the effects are more obvious in the medium and high dose groups than in the low dose group.
