摘要
目的:探讨银屑病患者血清对真皮间充质干细胞(DMSCs)免疫功能的影响。方法:用银屑病患者血清作用于正常DMSCs,采用实时荧光定量聚合酶链反应(real-time-PCR)检测IL-10、PGE_(2)、TLR4的mRNA表达水平,同时检测作用后的DMSCs对PHA活化的T细胞增殖的抑制能力。结果:银屑病患者血清作用后的DMSCs表达IL-10、PGE_(2)、TLR4的mRNA水平较正常对照组降低,且对PHA活化的T细胞增殖的抑制作用减弱,差异具有统计学意义(P<0.05)。结论:银屑病患者血清作用于DMSCs后,其血清中升高的炎症因子会导致DMSCs表达的TLR4、PGE_(2)、IL-10减少,进而降低间充质干细胞的免疫调节功能。
Objective:To investigate the effect of serum on the immune function of dermal mesenchymal stem cells(DMSCs)in patients with psoriasis.Methods:The normal dermal mesenchymal stem cells were treated with psoriatic serum,expression levels of IL-10,PGE_(2),TLR4 mRNA were detected by real-time-PCR,at the same time,the inhibition of DMSCs on the proliferation of PHA activated T cells was detected.Results:The mRNA expression levels of IL-10,PGE_(2) and TLR4 in the dermal mesenchymal stem cells treated with serum in psoriasis were lower than that in the normal controls,the inhibition of DMSCs on the proliferation of PHA activated T cells was weakened(P<0.05).Conclusion:After the serum of patients with psoriasis acts on the dermal mesenchymal stem cells,the increased inflammatory factors in their serum will lead to the decrease of TLR4,PGE_(2) and IL-10 expression of mesenchymal stem cells and then reduce the immunoregulatory function of mesenchymal stem cells.
