摘要
为了确诊一起疑似禽腺病毒(FAdV)引起的感染鸡临床病例,本试验对山东临沂某养殖场送检的病鸡进行剖检、病毒分离鉴定以及对分离到的病毒序列进行比对与同源性分析。结果发现:病鸡出现心包积液,肝脏有大小不等点状出血斑,肝脏颜色变浅等症状。处理后的病料经SPF鸡胚卵黄囊接种后,病毒可使鸡胚发育迟缓,胚体出血,肝脏出现白色坏死斑。对分离病毒的基因进行PCR扩增测序后,将获得的核苷酸序列与GenBank中公布的FAdV-11毒株的核苷酸序列进行对比,同源性为97%~100%。最后确诊该病原体为禽腺病毒Ⅰ群D11型。该研究结果为探究我国FAdV的分子流行病学与防控提供了基础材料和理论依据。
In order to diagnose a clinical case of fowl adenovirus(FAdV)infection on a poultry fl ock in Linyi city of Shandong province,pathological examination,virus isolation and identifi cation,sequence alignment and homology analysis were conducted in the present study.The diseased chickens had pericardial effusion,punctate hemorrhagic spots of varying sizes in the liver,and light liver color.Inoculation in yolk sacs of SPF chicken embryos caused retardation,hemorrhage and white necrotic spots in the livers of the inoculated embryo bodies.PCR amplification and sequencing analysis confirmed the isolated virus to be FAdV GroupⅠD11 as its nucleotide sequence homology was 97%-100%to the known nucleotides of the FAdV-11 strain published in GenBank.This study provided essential materials and information for further exploring the molecular epidemic situation and control of FAdV in China.
