摘要 目的基于网络药理学的方法探讨如意金黄散治疗疖肿的作用机制。方法通过中药系统药理学数据库(TCMSP)并检索先关文献来获得如意金黄散所含中药的主要化学成分,并收集其相对应的靶点;疖肿的疾病靶点来源于GeneCards、DrugBank、Home-OMIM-NCBI数据库。结合Cytoscape3.7.1构建药物-核心靶点-有效成分网络,靶蛋白相互作用(PPI)网络。利用Bioconductor中的R包对核心靶标进行基因本体(GO)功能富集和KEGG通路分析。结果根据筛选条件获得药物活性化学成分126个,药物靶点279个;获得疾病靶点165个;获得核心靶标20个;PPI蛋白作用网络分析,核心蛋白涉及表皮生长因子受体(EGFR)、白细胞介素(IL)-6、IL-1B等;GO功能富集分析发现主要参与了白细胞-细胞黏附、蛋白质定位建立的正调控、多细胞组织内稳态等生物学过程;KEGG通路主要涉及IL-17信号通路、AGE-RAGE信号通路、以及肿瘤坏死因子(TNF)信号通路等。结论如意金黄散治疗疖肿是多靶点、多途径的,本次研究为进一步研究其作用机制提供依据。 Objective In order to explore the mechanism of Ruyi Jinhuang powder in the treatment of furuncle based on the method of network pharmacology.Methods The main chemical components of traditional Chinese medicine(TCM)contained in Ruyi Jinhuang powder were obtained by TCM system pharmacology database(TCMSP),and the corresponding targets were collected;the disease targets of furuncle were from GeneCards,DrugBank,Home-OMIM-NCBI.Combined with Cytoscape 3.7.1,a PPI network of drug core target active component was constructed.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis were carried out for the core target using the R-packet in Bioconductor.Results According to the screening conditions,126 active chemical components and 279 drug targets were obtained;165 disease targets were obtained;20 core targets were obtained;PPI protein network and network topology were analyzed,and the core proteins involved EGFR,IL-6,IL-1B,and so on.GO function enrichment analysis found that it was mainly involved in leukocyte cell adhesion,positive regulation of protein location establishment,and multicellular tissue homeostasis KEGG pathway was mainly involved in interleukin(IL)-17 signaling pathway,AGE-RAGE signaling pathway,and tumor necrosis factor(TNF)signaling pathway.Conclusion Ruyi Jinhuang powder is multi-target and multi-channel in the treatment of furuncle.This study provides basis for further study of its mechanism.
