HPLC法同时测定复方紫草微乳凝胶中三种成分的含量

摘要 目的建立HPLC法同时测定复方紫草微乳凝胶中阿魏酸、欧前胡素、异欧前胡素含量的方法。方法采用Hypersil BDS C_(18)(250 mm×4.6 mm,5μm)色谱柱,流动相甲醇(A)-0.1%磷酸水溶液(B)梯度洗脱,流速1.0 mL·min^(-1),柱温35℃,检测波长300 nm。结果阿魏酸、欧前胡素、异欧前胡素线性范围分别为0.17075~2.736μg(r=0.9998)、0.528~8.448μg(r=0.9998)、0.2275~3.64μg(r=0.9997);平均回收率(n=6)分别为98.46%、99.80%、100.23%,RSD分别为1.78%、0.78%、1.63%。含量测定结果(n=3)分别为0.7348~0.7609、2.5887~2.6709、1.1391~1.1589 mg·g^(-1)。结论该方法为复方紫草微乳凝胶的质量标准研究提供参考。 OBJECTIVE To develop an HPLC method for simultaneously determining 3 components including ferulic acid,imperatorin and isoimperatorin in compound Arnebiae radix microemulsion gel.METHODS The Hypersil BDS C_(18)(250 mm×4.6 mm,5μm)was adopted.The mobile phase was composed of methanol(A)-0.1%aqueous phosphoric acid(B)in a linear gradient elution mode.The flow rate was 1.0 mL·min^(-1).The column temperature was 35℃.The detection wavelength was set at 300 nm.RESULTS The linear ranges of ferulic acid,imperatorin and isoimperatorin fell within the ranges of 0.17075-2.736μg(r=0.9998)、0.528-8.448μg(r=0.9998)、0.2275-3.64μg(r=0.9997),respectively.The recoveries(n=6)of ferulic acid,imperatorin and isoimperatorin were 98.46%、99.80%、100.23%,separately,and the relative standard deviations were 1.78%、0.78%、1.63%,respectively.The results of content assay(n=3)were 0.7348-0.7609 mg·g^(-1)、2.5887-2.6709 mg·g^(-1)、1.1391-1.1589 mg·g^(-1),separately.CONCLUSION This method can provide the reference for quality standard for compound Arnebiae radix microemulsion gel.
