一测多评法测定乌蕨中5个黄酮成分含量 被引量:11

Determination of 5 flavonoids in Sphenomeris chinensis by quantitative analysis of multi-components by single marker

摘要 目的:建立一测多评法测定乌蕨中5个黄酮成分的含量,并验证其准确性及可行性。方法:采用高效液相色谱法,用牡荆素作内参物,计算荭草苷、牡荆素葡萄糖苷、牡荆素鼠李糖苷、木犀草苷的相对校正因子,用相应的校正因子计算各组分的含量,实现一测多评。同时用外标法测定乌蕨中5个黄酮成分含量,通过结果比较,验证一测多评法的准确性及可行性。结果:在一定线性范围内,牡荆素与荭草苷、牡荆素葡萄糖苷、牡荆素鼠李糖苷、木犀草苷的相对校正因子分别为0.772、1.368、1.380、0.731。用一测多评法测得27批样品中4个黄酮成分的含量与外标法测定结果无显著性差异。结论:建立乌蕨中荭草苷、牡荆素葡萄糖苷、牡荆素鼠李糖苷、牡荆素、木犀草苷含量的一测多评法方法可行,测定结果准确、可信,可用于乌蕨药材及饮片定量分析及质量控制。 Objective:To establish the quantitative analysis of multi-components by single marker(QAMS)method for the content determination of five flavonoids in Sphenomeris chinensis,and verify its accuracy and feasibility.Methods:The relative correction factors of orientin,glucosyl-vitexin,vitexin-2-o-rhamnoside and galuteolin were calculated by HPLC with vitexinas internal reference,and the contents of each component were calculated with the corresponding correction factors,so as to realize quantitative analysis of multi-components by single marker.At the same time,the contents of five flavonoids in sphenomeris chinensis weredetermined by external standard method,and the accuracy and feasibility of QAMSmethod were verified by comparing the results.Results:Within a certain linear range,the relative correction factors between vitexin and orientin,glucosyl-vitexin,vitexin-2-o-rhamnoside as well as galuteolin were 0.772,1.368,1.380 and 0.731,respectively.There was no significant difference betweenthe contents of 4 flavonoids in 27 batches of samples determined by twomethods.Conclusion:The quantitative analysis of multi-components by single marker method for the contentdetermination of orientin,glucosyl-vitexin,vitexin-2-o-rhamnoside,vitexin and galuteolin is feasible,accurate and reliable,which can be used for quantitative analysis and quality control of sphenomeris chinensis and its decoction pieces.
