摘要
海藻酸在食品、医药、材料等领域有着广泛的用途,从海藻中提取是目前海藻酸的主要制备方法。本研究以提高马尾藻(Sargassum)中海藻酸的提取率为目标,系统研究了利用纤维素酶、果胶酶和海藻酸裂解酶复合提取马尾藻中海藻酸工艺。实验探究了酶添加量、时间、温度和pH对提取海藻酸的影响,并通过正交试验进一步优化提取工艺。结果表明在料液比1∶20(m/v),添加2%的纤维素酶、2%的果胶酶和2%的海藻酸裂解酶,提取温度55℃、提取pH 4.5,酶解90 min后再采用钙析-酸化法制备海藻酸的工艺条件下,海藻酸提取率可达25.43%,相对于传统化学法提高了163%,相对于市场上现有商品复合酶提取法提高了20%。复合酶法提取海藻酸的酶解效率高、海藻酸产品品质好,为复合酶法从马尾藻原料中高效提取海藻酸提供了数据支撑。[中国渔业质量与标准,2023,13(2):18-24].
Alginate is widely used in food,medicine,materials and other fields,and extracting it from seaweed is the main method for preparing alginate.In order to improve the extraction rate of alginate from Sargassum,the extraction process of alginate from Sargassum by using cellulase,pectinase and alginate lyase was systematically studied.The effects of enzyme amount,time,temperature and pH on the extraction of alginate were investigated,and the extraction process was further optimized by orthogonal test.The results showed that 2%cellulase,2%pectase and 2%alginate lyase were added in the solid-liquid ratio of 1∶20,the extraction temperature 55℃,pH 4.5.After enzymatic digestion for 90 min,alginate was prepared by calcium analysis-acidification method,and the extraction rate can reach 25.43%,which is 163%higher than the traditional chemical method and 20%higher than the other commodity compound enzymes extractionmethod in the market.Compound enzymes extraction method has the high enzymatic efficiency and high product quality,and it provides data support for the efficient extraction of alginate from the Sargassum.[Chinese Fishery Quality and Standards,2023,13(2):18-24]
