黄芩总黄酮部位对常见水产致病菌的体外抑制作用

The Inhibitory Effect of the Total Flavonoid Fractions from Scutellaria Baicalensis Georgi on Common Aquatic Pathogenic Bacteria

摘要 为寻找水产养殖领域天然、安全的抗生素替代品,探索黄芩醇提物对水产致病菌抑制作用的总黄酮部位.采用大孔树脂分离和抑菌圈法确定黄芩抗菌的总黄酮部位,采用紫外分光光度法和HPLC法分别对其进行定量和定性分析,采用琼脂稀释法测定黄芩总黄酮部位对7种水产致病菌的MIC.研究结果表明,黄芩中的黄酮类成分主要富集在30%醇洗和70%醇洗部位,并表现出明显的抑菌活性,其总黄酮含量达75.2%.黄芩总黄酮部位对其中6种水产致病菌的10个菌株均表现出较强的抑制作用,其MIC在0.36~1.43 mg/mL之间.黄芩总黄酮部位具有较广谱的抗水产致病菌作用,在水产养殖业有着广泛的应用前景. In order to address a natural and safe alternative drug to avoid abuse of antibiotics in the aquaculture field,the total flavone fraction of ethanol extract of Scutellaria baicalensis Georgi(SG)was studied.The results showed that the total flavonoid fractions of the ethanol extract of SG had strong inhibitory effects on the aquatic pathogenic bacteria.The total flavonoid fractions of SG were isolated by macroporous resin separation and identified by using the inhibition zone method.Next,the total flavonoid fractions were then qualitatively and quantitatively analyzed by UV spectrophotometry and HPLC,respectively.The minimum inhibitory concentration(MIC)of the total flavonoid fractions of SG against 7 aquatic pathogens was determined by agar dilution method.The results showed that the flavonoids in the extract of SG were mainly concentrated in 30%and 70%alcohol elution,showing obvious antibacterial activity,and the total flavonoid content reached 75.2%.The total flavonoid fractions of SG showed the strong inhibition on the 10 strains from 6 aquatic pathogenic bacteria,and their MIC values were 0.36~1.43 mg/mL.Therefore,the total flavonoid fraction of SG has broad-spectrum antibacterial activity against aquatic pathogens,and shows a potential anti-bacterium application in aquaculture.
