摘要 目的分析2021年从福建省发生腹泻的猪场分离到的1株G26P[23]新基因型猪轮状病毒RVA/pig/CHN/FJSH01/2021/G26P[23](简称FJSH01)的分子特征。方法利用MARC-145细胞进行RV分离,采用RT-PCR方法对分离株FJSH01的全基因组进行了测定,利用在线分型工具RotaC v2.0对其基因型进行鉴定,并对其分子遗传特征进行了分析。结果分离株FJSH01的基因型图谱为G26-P[23]-I5-R1-C1-M1-A8-N1-T1-E1-H1,即G26P[23]型。分离株FJSH01的VP3、NSP1、NSP3和NSP5基因分别与人源轮状病毒RVA/Human_wt/VNM/30378/2009/G26P[19]、RVA/Human-wt/VNM/NT0077/2007/G4P[6]、Human/LL4260/China/T1和RVA/Human/Ryukyu-1120的核苷酸同源性最高,为96.01%~98.65%,其余7个片段与猪源轮状病毒(PoRV)的核苷酸同源性最高,为95.12%~97.99%,表明分离株FJSH01可能为人源轮状病毒和猪源轮状病毒重配后的病毒株。结论G26P[23]型PoRV为国内首次鉴定,研究结果丰富了PoRV分子流行病学资料,为PoRV预防控制提供理论参考。 To understand the genetic characteristics of a new genotype G26P[23]porcine rotavirus(PoRV)RVA/pig/CHN/FJSH01/2021/G26P[23](referred to as FJSH01)isolated from piglets with diarrhea in Fujian Province in 2021,we sequenced its complete genome and analyzed its molecular genetic characteristics.Samples were inoculated into MARC-145 cells for PoRV isolation,and 11 segments of FJSH01 were amplified by RT-PCR.The genotype constellation of FJSH01 was identified with an RotaC v2.0 automated online genotyping tool.The genotype constellation of FJSH01was G26-P[23]-I5-R1-C1-M1-A8-N1-T1-E1-H1(G26P[23]genotype).Sequence analysis indicated that the VP3,NSP1,NSP3 and Nsp5 genes of FJSH01 had the highest sequence identities(96.01%-98.65%)with respect to human group A rotavirus(RVA/Human_wt/VNM/30378/2009/G26P[19],RVA/Human-wt/VNM/NT0077/2007/G4P[6],Human/LL4260/China/T1 and RVA/Human/Ryukyu-1120).The remaining seven segments of FJSH01 had the highest sequence identities(95.12%-97.99%)with PoRV.The above results suggested that FJSH01 may be a reassortant virus between human and porcine rotavirus.In conclusion,the G26P[23]genotype PoRV was first identified in China,and the results of this study provide molecular epidemiological data on PoRV that may serve as a theoretical reference for the prevention and control PoRV.
