基于^(1)H NMR代谢组学技术研究壮医经筋疗法治疗单纯性肥胖的作用

摘要 目的:基于^(1)H NMR代谢组学方法探讨壮医经筋疗法治疗单纯性肥胖血清代谢组学的相关代谢机制。方法:将单纯性肥胖志愿受试者纳入治疗组,健康人群纳入空白组,治疗组采用壮医经筋推拿、针刺及拔罐治疗,空白组不采取任何治疗,于治疗前后采集治疗组血清及空白组血清,应用^(1)H NMR的代谢组学方法对各组受试者的血清代谢组进行分析,寻找各组间的差异代谢标志物。结果:采用正交投影偏最小二乘法判别分析(OPLS-DA),以变异权重系数(VIP)>1、P<0.05和FC>2为限定条件对所构建的数学模型进行潜在生物标志物的筛选,共筛选出潜在生物标志物11种,分别为异亮氨酸、脯氨酸、牛磺酸、半乳糖、缬氨酸、柠檬酸盐、苹果酸盐、半胱氨酸、甘露醇、葡萄糖醇和丙氨酸。采用MetaboAnalyst 5.0对标志物涉及的代谢通路进行归属注释和富集分析,得到与疾病相关的关键代谢通路为牛磺酸和亚牛磺酸代谢通路及三羧酸循环。结论:壮医经筋疗法可能通过调节异亮氨酸、脯氨酸、牛磺酸、半乳糖、缬氨酸、柠檬酸盐、苹果酸盐、半胱氨酸、甘露醇、葡萄糖醇和丙氨酸等物质,改善牛磺酸、亚牛磺酸代谢通路和三羧酸循环等代谢通路,以达到治疗单纯性肥胖的目的。 Objective:To explore the mechanism of serum metabolomics in simple obesity treated by meridian sinew therapy of Zhuang medicine based on^(1)H NMR metabolomics.Methods:The volunteers with simple obesity were included in the treatment group,and the healthy people were included in the blank group.The treatment group was treated with meridian sinew Tuina massage of Zhuang medicine,acupuncture and cupping,and the blank group did not receive any treatment.The serum were collected before and after the treatment.The serum metabolomics of each group was analyzed by^(1)H NMR metabolomics method to find the differential metabolic markers between the two groups.Results:Orthogonal partial least-squares discriminnation analysis(OPLS-DA)was used to screen the potential biomarkers of the established mathematical model,with variation weighting coefficient(VIP)>1,P<0.05 and FC>2 as the limiting conditions.A total of 11 potential biomarkers were screened,including isoleucine,proline,taurine,galactose,valine,citrate,malate,cysteine,mannitol,glucitol and alanine.MetaboAnalyst 5.0 was used to perform attribution annotation and enrichment analysis on the metabolic pathways involved in the markers,and the key metabolic pathways related to the disease were taurine and hypotaurine metabolic pathways and the tricarboxylic acid cycle.Conclusion:Meridian sinew therapy of Zhuang medicine may improve the metabolic pathways of taurine and hypotaurine and the metabolic pathways of tricarboxylic acid cycle by regulating substances including isoleucine,proline,taurine,galactose,valine,citrate,malate,cysteine,mannitol,glucose alcohol and alanine,so as to achieve the purpose of treating simple obesity.
