思肤安营养修护仿生膏对痤疮大鼠皮损及IGF-1/PI3K/Akt信号通路的影响

摘要 目的 观察思肤安营养修护仿生膏对痤疮大鼠皮损的影响,并基于胰岛素样生长因子1(IGF-1)/磷脂酰肌醇-3-激酶(PI3K)/蛋白激酶B(Akt)信号通路探究其作用机制。方法将18只SD大鼠随机分为对照组、阿达帕林组及思肤安组,每组6只。3组均采用痤疮丙酸杆菌注射诱导建立痤疮模型,造模成功后,阿达帕林组及思肤安组分别给予阿达帕林凝胶和思肤安营养修护仿生膏外涂,对照组给予生理盐水外涂,均1次/d,连续干预4周。分别于干预2周及4周后观察3组大鼠造模区域皮损及HE染色组织病理变化,尼罗河染色观察皮损组织皮脂分泌情况,Western blot及免疫组化法检测皮损组织中IGF-1、PI3K、Akt、甾醇调节元件结合蛋白1(SREBP-1)蛋白表达情况。结果 干预2周后,思肤安组、阿达帕林组痤疮鼠背部皮损处结节、红肿较对照组轻,毛漏斗内角质物、毛囊壁及周围组织炎细胞浸润均较对照组减少;干预4周后,思肤安组及阿达帕林组丘疹、结节基本消退,表皮各层结构组织基本正常,其中思肤安组皮损恢复更好。思肤安组皮损脂质百分比呈现先上升后下降的趋势,干预4周后脂质百分比明显高于阿达帕林组(P<0.05),与阿达帕林组干预2周后相当(P>0.05)。干预2周、4周后阿达帕林组和思肤安组皮损组织中IGF-1、PI3K、Akt、SREBP-1蛋白相对表达量均明显低于对照组(P均<0.05),且思肤安组均明显低于阿达帕林组(P均<0.05),免疫组化染色也显示干预2周、4周后阿达帕林组和思肤安组皮损组织中IGF-1、PI3K、Akt、SREBP-1蛋白表达均较对照组明显减少。结论 思肤安营养修护仿生膏有一定的脂质调节作用,可通过抑制IGF-1/PI3K/Akt通路下调SREBP-1的表达发挥治疗痤疮作用。 Objective It is to observe the effect of Safecare Bionic Trentment Skin Cream on skin lesions of rats with acne,and to explore its mechanism of action based insulin-like growth factor(IGF-1)/phosphatidylinositol-3-kinase(PI3K)/protein kinase B(Akt)signal pathway.Methods Eighteen SD rats were randomly divided into control group,adaparine group and Safecare group,with 6 rats in each group.All three groups were injected with acnes Propionibacterium to establish acne models,and after successful modeling,the adapalene group and Safecare group were given adapalene gel and Safecare Bionic Trentment Skin Cream by external application,and the control group was given normal saline by external application,all once daily,continuously treated for 4 weeks.The changes of skin lesions and histopathology of HE staining in the modeling area of of rats of the three groups were observed after 2 and 4 weeks of treatment,and the sebum secretion of skin lesions was observed by Nile staining,the protein expressions of IGF-1,PI3K,Akt,and sterol regulatory element binding protein 1(SREBP-1)in the skin lesions was detected by Western blot and immunohistochemistry.Results After 2 weeks of treatment,the nodules,red swelling at the back skin lesions of acne rats in the Safecare group and adapalene group were lighter than those in the control group,and the keratinous substance in the follicular infundibulum,infiltration of inflammatory cells in the follicles walls and its surrounding tissues were reduced compared with those in the control group;after 4 weeks of treatment,the pimples and nodules basically disappeared and the structural tissues of all layers of the epidermis were basically normal in the Safecare group and adapalene group,and the recovery of the lesions in the Safecare group was better.The sebum percentage of skin lesions in the Safecare group showed a trend of increasing and then decreasing,and the sebum percentage was significantly higher than that in the adapalene group after 4 weeks of treatment(P<0.05),and was equal to that in the adapalene group after 2 weeks of treatment(P>0.05).The relative expressions of IGF-1,PI3K,Akt,and SREBP-1 proteins in the lesion tissues of the adapalene group and Safecare group were significantly lower than those of the control group after 2 and 4 weeks of treatment(all P<0.05),and the expressions of the Safecare group were significantly lower than those of the adapalene group(all P<0.05),immunohistochemical staining also showed that the expressions of IGF-1,PI3K,Akt,and SREBP-1 in the lesion tissues of the adapalene and Safecare groups were significantly higher than those of the control group after 2 and 4 weeks of treatment(all P<0.05).Conclusion Safecare Bionic Trentment Skin Cream has a certain effect of sebum regulating,and can down-regulate the expression of SREBP-1 to treat acne via inhibiting IGF-1/PI3K/Akt pathway.
