女贞子、α-甘露聚糖肽对热应激小鼠血清激素水平及肝脏热应激蛋白表达的影响 被引量:1

摘要 试验旨在观察小鼠热应激模型的血清激素水平和肝脏热应激蛋白激素的动态水平,探讨女贞子、α-甘露聚糖肽抗热应激的调节作用。选取日龄、体型、体重相近的40只小白鼠,随机分为5组,每组8重复,每个重复1只,分别为空白组、热应激对照组、女贞子组(0.2 mL)、α-甘露聚糖肽组(0.2 mL)、女贞子+α-甘露聚糖肽组(各0.1 mL)。空白对照组小鼠正常饲养,其他4组均进行热应激处理。结果表明,当小鼠受到热应激,单独使用女贞子和α-甘露聚糖肽可以使小鼠血糖(GLU)、肌酸激酶(CK)、乳酸脱氢酶(LDH)、皮质醇(CORT)水平极显著降低(P<0.01)。当两者混合使用时,小鼠GLU、LDH、CORT水平与热应激对照组差异不显著(P>0.05),CK活性极显著高于热应激对照组(P<0.01)。当小鼠处于热应激状态时,热休克蛋白HSP60、HSP70、HSP90的表达量分别是空白组的1.91、2.03、1.26倍,表达量明显提高。单独使用女贞子和α-甘露聚糖肽后,HSP60、HSP90的表达量明显低于热应激对照组,HSP70的表达量相比空白组和热应激对照组均明显上升。当两种药物同时使用时,HSP60、HSP90的表达量和热应激对照组相比无明显差异,HSP70的表达量明显高于热应激对照组。研究表明,女贞子和α-甘露聚糖肽在单独使用时能够显著降低小鼠的热应激反应程度,两种药物混合使用可能无缓解热应激作用或者两药物相互拮抗。 The experiment was to investigate the regulatory effect of Ligustrum lucidum andα-mannan peptide on serum hormone level and liver heat stress protein expression of heat stress mice.A total of 40 mice with similar age,body size and body weight were randomly divided into five groups,with eight replicates in each group and one mice in each replicate,which were divided into blank group,heat stress control group,normal Ligustrum lucidum group(0.2 mL),α-mannan peptide group(0.2 mL)and Ligustrum lucidum+α-mannan peptide group(0.1 mL each).The mice in blankgroup were fed normally,and the mice in other four groups were treated with heat stress.The results showed that when mice were subjected to heat stress,the levels of blood glucose,creatine kinase,lactate dehydrogenase and cortisol of mice were extremely decreased after using Ligustrum lucidum andα-mannan peptide alone(P<0.01).There was no significant difference in blood glucose concentration,lactate dehydrogenase and cortisol contents between the Ligustrum lucidum+α-mannan peptide group and heat stress control group(P>0.05),and the activity of creatine kinase of Ligustrum lucidum+α-mannan peptide group was extremely higher than that of heat stress control group(P<0.01).When the mice were under heat stress,the relative expression levels of HSP60,HSP70 and HSP90 were 1.91,2.03 and 1.26 times of those in blank group,respectively,and the expression levels were significantly increased.Using Ligustrum lucidum andα-mannan peptide alone,the expression levels of HSP60 and HSP90 were significantly lower than those of heat stress control group,while the expression levels of HSP70 were significantly increased compared to blank group and heat stress control group.The expression of HSP70 was significantly increased compared with blank control group and heat stress control group.When both drugs were used simultaneously,there was no significant difference in the expression levels of HSP60 and HSP90 compared to heat stress control group,while the expression level of HSP70 was significantly higher than that of the heat stress control group.The study indicates thatLigustrum lucidum andα-mannan peptide can significantly reduce the degree of heat stress response in mice when used alone,but the combination of the two drugs may not alleviate heat stress or antagonize each other.
