TGF-β/Smad通路在羊栖菜多糖诱导肝癌细胞凋亡过程中的作用

摘要 目的:探讨羊栖菜多糖通过转化生长因子β(TGF-β)/Smad信号通路对肝癌细胞增殖、凋亡和侵袭迁移的影响。方法:不同浓度羊栖菜多糖处理对数生长期的HepG2细胞,CCK-8法检测细胞存活率,流式细胞仪检测细胞凋亡率,Transwell法检测细胞侵袭和迁移能力,蛋白印迹法检测TGF-β1、p-Smad2、B淋巴细胞瘤-2基因(Bcl-2)、Bcl-2相关X蛋白(Bax)相对表达量。结果:细胞存活率、迁移和侵袭细胞数以及TGF-β1、p-Smad2和Bcl-2蛋白相对表达量随羊栖菜多糖浓度的升高而降低,细胞凋亡率和Bax蛋白相对表达量随羊栖菜多糖浓度的升高而升高(P<0.05)。结论:羊栖菜多糖可抑制肝癌细胞增殖、迁移和侵袭,诱导癌细胞凋亡,可能是通过抑制TGF-β/Smad信号通路发挥作用。 Objective:To investigate the effects of sargassum fusiforme on the proliferation,apoptosis,invasion,and migration of hepatocarcinoma cells through transforming growth factorβ(TGF-β)/Smad signaling pathway.Methods:The hepG2 cells at logarithmic growth stage were treated with different concentrations of sargassum fusiforme.Cell survival rate was detected by CCK-8 method,cell apoptosis rate was detected by flow cytometry,cell invasion and migration ability was detected by the Transwell method,and TGF-β1,p-Smad2,and B lymphocytoma-2(Bcl-2)genes were detected by western blot.Relative expression of Bcl-2 associated X protein(Bax).Results:The cell survival rate,the number of migrating and invading cells and the relative expression levels of TGF-β1,p-Smad2 and Bcl-2 proteins were decreased with the increase of sargassum fusiforme concentration,while the apoptosis rate and the relative expression levels of Bax protein were increased with the increase of sargassum fusiforme concentration(P<0.05).Conclusion:Sargassum fusiforme can inhibit the proliferation,migration,and invasion of hepatocarcinoma cells and induce apoptosis of cancer cells,possibly by inhibiting TGF-β/Smad signaling pathway.
