瓦草中的化学成分及其抗炎活性研究

摘要 目的研究瓦草中的化学成分及其抗炎活性。方法应用硅胶、ODS及高效液相等多种色谱法对提取物进行分离纯化,并通过核磁共振和质谱技术鉴定化合物的结构。采用环氧合酶-2(COX-2)试剂盒对分离得到的单体化合物进行抗炎活性筛选。结果从瓦草的70%乙醇提取物中分离鉴定了8个单体化合物,包括5个三萜皂苷类化合物[sinocrassulosideⅦ(1)、sinocrassulosideⅥ(2)、sinocrassulosideⅨ(3)、sinocrassulosideⅧ(4)、sinocrassulosideⅩ(5)];2个蜕皮激素类化合物[蜕皮激素(6)、蜕皮激素-3-O-α-D-甘露糖(7)]及1个咔伯啉生物碱类化合物[l-乙酰基-3-甲酯基-β-咔啉(8)],其中,化合物7为新化合物,化合物8为首次从蝇子草属植物中分离得到。化合物3和7的抗炎活性较强,IC50值分别为9.36μmol·L^(-1)和6.70μmol·L^(-1)。结论瓦草化学成分具有多样性,且分离出来的单体成分均具有一定的抗炎活性。 Objective To determine the chemical constituents and anti-inflammatory activities of Silene viscidula.Methods The extracts were isolated and purified by silica gel,ODS and HPLC,and their structures were identified by NMR and MS.The anti-inflammatory activity of those constituents was screened by COX-2.Results Totally 8 compounds were isolated and identified from 70%ethanol extract of Silene viscidula,including five triterpenoid saponins:sinocrassulosideⅦ(1),sinocrassulosideⅥ(2),sinocrassulosideⅨ(3),sinocrassulosideⅧ(4),and sinocrassulosideⅩ(5);two ecdystin compounds:hydroxyecdysone(6),hydroxyecdysone-3-O-α-D-mannose(7);one carboline alkaloid compound:1-acetyl-3-methoxycarbonyl-β-carboline(8).Compound 7 was a new compound and compound 8 was first isolated from the genus of Silene.Compounds 3 and 7 had strong anti-inflammatory activities,with IC50 values of 9.36μmol·L^(-1) and 6.70μmol·L^(-1).Conclusion There are various chemical constituents in Silene viscidula,and the isolated compounds show certain anti-inflammatory activity.
