SPE-HPLC法测定31批次鱼中孔雀石绿残留量

摘要 目的:建立SPE-HPLC测定31批次鱼中孔雀石绿残留的方法。方法:ZORBAX SB-C_(18)柱(4.6 mm×250 mm,5μm),流动相为乙腈-乙酸铵缓冲溶液(0.125 mol·L^(-1),pH=4.5),梯度洗脱,激发波长265 nm,发射波长360 nm,柱温35℃,进样量20μL,流速1.3 mL·min^(-1)。结果:孔雀石绿在0.005~0.200μg·mL^(-1)线性关系良好,相关系数R=0.99961;回收率在82.4%~84.2%,RSD为0.82%。结论:该方法快捷简便、准确高效,适用于鱼中孔雀石绿残留量的检测。 Objective:To establish a SPE-HPLC method for the determination of malachite green residues in 31 batches of fish.Method:The chromatographic column was ZORBAX SB-C_(18) column(4.6 mm×250 mm,5μm).The mobile phase was acetonitrile-ammonium acetate buffer solution(0.125 mol·L^(-1),pH=4.5)with gradient elution.The excitation wavelength was 265 nm,the emission wavelength was 360 nm,the column temperature was 35℃,the injection volume was 20μL,and the flow rate was 1.3 mL·min^(-1).Result:Malachite green showed a good linear relationship in the range of 0.005~0.200μg·mL^(-1),the correlation coefficient R=0.99961;the recovery rate ranges from 82.4%to 84.2%,with an RSD of 0.82%.Conclusion:The method is fast,simple,accurate and efficient,which is suitable for the detection of malachite green residues in fish.
