高温处理对补骨脂浸膏中酪氨酸酶活性成分的影响

摘要 采用高效液相色谱法同时测定高温处理后补骨脂中补骨脂素、异补骨脂素、新补骨脂素异黄酮、补骨脂二氢黄酮用甲醚、大豆苷元、补骨脂甲素和补骨脂乙素等七种成分的含量变化,及采用体外酪氨酸酶多巴速率氧化微量法测定七种成分的酪氨酸酶活性,评价高温处理对补骨脂中酪氨酸酶活性成分的影响.结果显示,与高温处理前相比,高温处理后补骨脂中7种成分的含量发生了变化;在补骨脂注射液100℃30 min条件下,除大豆苷元的含量最高外,其他6种成分的含量均不是最高,但是补骨脂素和异补骨脂素的含量下降最多;补骨脂素和异补骨脂素在浓度较低时对酪氨酸酶具有抑制作用,其他5种成分对酪氨酸酶均有不同程度的激活作用.可见,高温处理对补骨脂中酪氨酸酶活性成分具有一定的影响,该研究可为补骨脂的进一步研究提供参考与依据. An HPLC method was used for simultaneous determination of seven components including psoralen,isopsoralen,neobavaisoflavone,bavachinin,daidzein,corylifolin,corylifolinin from Psoralea corylifolia after high-temperature treatment.The oxidation rate of L-DOPA in vitro was measured to detect tyrosinase activity,for evaluating the effect of high temperature treatment on tyrosinase active components in P.corylifolia.The results showed that the contents of seven components were changed after high temperature treatment compared with that before high temperature treatment.At the condition 100℃30 min,in addition to the highest content of daidzein,the contents of other six components were all not the highest,but the contents of psoralen and isopsoralen were the biggest drops,and psoralen and isopsoralen had the effects of inhibiting tyrosinase at low concentrations,other five components had different degree of tyrosinase activation.High temperature treatment had certain effect on the active constituents of P.corylifolia.This study could provide reference and basis for further research on P.corylifolia.
