经典名方二冬汤物质基准高效液相色谱指纹图谱建立

HPLC Fingerprint of Classic Famous Formulas Erdong Decoction Substance Reference

目的建立经典名方二冬汤物质基准的高效液相色谱(HPLC)指纹图谱。方法色谱柱为Inertsil ODS-4柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.05 mol/L乙酸铵溶液-冰醋酸(梯度洗脱),流速为1.0 mL/min,检测波长为254 nm,柱温为30℃,进样量为10μL。采用中药色谱指纹图谱相似度评价系统(2012版)建立二冬汤物质基准HPLC指纹图谱,指认和归属共有峰,并对15批样品进行相似度评价。结果15批样品的对照指纹图谱的相似度均不低于0.992;建立了9个共有峰的指纹图谱,指认芒果苷(峰1)、金丝桃苷(峰3)、黄芩苷(峰4)、千层纸素A-7-O-β-D-葡萄糖醛酸苷(峰6)、汉黄芩苷(峰7)、甘草酸(峰8)、黄芩素(峰9)7个共有峰;归属峰1为知母色谱峰,峰2、峰3为荷叶色谱峰,峰4、峰5、峰6、峰7、峰9为黄芩色谱峰,峰8为甘草色谱峰。结论所建立的方法易操作,结果可靠,可用于二冬汤物质基准的质量控制。

Objective To establish a high-performance liquid chromatography(HPLC)fingerprint of the classic famous formula Erdong Decoction substance reference.Methods The chromatographic column was Inertsil ODS-4 column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-0.05 mol/L ammonium acetate solution-glacial acetic acid(gradient elution),the flow rate was 1.0 mL/min,the detection wavelength was 254 nm,the column temperature was 30℃,and the injection volume was 10μL.The HPLC fingerprint of Erdong Decoction Substance Reference was established by the Similarity Evaluation System of Chromatographic Fingerprint of Traditional Chinese Medicine(Version 2012),and common peaks were identified.The similarity of 15 batches of samples was evaluated.Results The similarity of the reference fingerprint of 15 batches of samples was higher than 0.992.The fingerprints of nine common peaks were established,and seven common chromatographic peaks were identified,including mangiferin(peak 1),hyperoside(peak 3),baicalin(peak 4),and oroxylin A-7-O-β-D-glucuronide(peak 6),wogonoside(peak 7),glycyrrhizic acid(peak 8),and baicalein(peak 9).Peak 1 was the chromatographic peak of Anemarrhenae Rhizoma,peaks 2 and 3 were the chromatographic peaks of Nelumbinis Folium,peaks 4,5,6,7,and 9 were the chromatographic peaks of Scutellariae Radix,and peak 8 was the chromatographic peak of Glycyrrhizae Radix et Rhizoma.Conclusion The established method is simple and reliable,which can be used for the quality control of Erdong Decoction substance reference.
