摘要 目的:探讨COQ10B对食管鳞癌细胞增殖、侵袭能力的影响及可能的分子机制。方法:通过TIMER2.0数据库分析COQ10B基因在各种肿瘤和相邻正常组织之间的表达水平。免疫组织化学检测COQ10B在食管鳞癌组织中的表达水平并进行临床病理特征分析及生存分析;利用慢病毒构建COQ10B敲减稳转株及Snail过表达稳转株,qRT-PCR和Western blot方法检测转染效率。MTT法、Celigo细胞计数及Transwell侵袭小室检测COQ10B对KYSE150细胞系增殖和侵袭能力的影响。通过GEPIA2数据库分析COQ10B表达水平与Snail的相关性;Western blot检测敲减COQ10B对Snail蛋白表达的影响。通过拯救实验分析COQ10B是否通过正调控Snail促进KYSE150细胞系增殖和侵袭能力。结果:COQ10B在ESCC组织及细胞系中均为高表达水平,且表达水平与临床病理特征病理分级相关。COQ10B与ESCC患者预后相关,高表达COQ10B的ESCC患者预后明显较差,且发现性别、肿瘤TNM分期是影响ESCC预后的独立危险因素。敲减COQ10B可降低ESCC细胞增殖及侵袭能力。COQ10B表达水平与Snail呈正相关,并通过正调控Snail促进KYSE150细胞系增殖和侵袭能力。结论:COQ10B通过正向调控Snail基因表达促进食管鳞癌增殖与侵袭。 Objective:To investigate the effect and possible molecular mechanism of COQ10B on cell proliferation and invasion of esophageal squamous cell carcinoma(ESCC).Methods:The expression level of COQ10B gene between various tumors and normal tissues was analyzed by TIMER2.0 database.The expression level of COQ10B in ESCC tissue was detected by immunohistochemistry.Clinicopathological features and survival were analyzed.Lentivirus were used to construct cell lines for stable COQ10B knockdown and Snail overexpression,and the transfection effect was detected by qRT-PCR and Western blot.The effect of COQ10B on cell proliferation and invasion was detected by MTT,Celigo cell count and Transwell invasion in KYSE150 cell line.The correlation between COQ10B expression level and Snail was analyzed by GEPIA2.The effect of COQ10B knockdown on Snail protein expression was analyzed by Western blot.Rescue experiments were conducted to analyze whether COQ10B promoted the proliferation and invasion ability of KYSE150 cell line by positively regulating Snail.Results:COQ10B was highly expressed in ESCC tissues and cell lines,and the expression level was correlated with pathological grade.COQ10B was associated with the prognosis of ESCC patients,and ESCC patients with high expression of COQ10B had a significantly worse prognosis,and gender and TNM stage were independent risk factors for ESCC prognosis.Knocking down COQ10B can reduce the proliferation and invasion ability of ESCC cells.The expression level of COQ10B was positively correlated with Snail,and COQ10B promoted the proliferation and invasion ability of KYSE150 cell line by positively regulating Snail.Conclusion:COQ10B can promote the proliferation and invasion of KYSE150 cells of esophageal squamous cell carcinoma through positive regulation of Snail gene expression.
