鸡白痢沙门氏菌自制抗原的制备及检测效果评价

摘要 为了评价鸡白痢沙门氏菌自制抗原的检测效果,试验从通过特异性凝集试验和非特异性凝集试验筛选结果中随机选择1株鸡白痢沙门氏菌临床分离株SP-41,与鸡白痢沙门氏菌标准菌株CVCC1792按照1∶1比例混合,采用平板凝集试验和微量凝集试验两种方法分别制备染色抗原和凝集抗原,并以100份鸡血清样品为检测对象,比较了自制抗原、鸡白痢鸡伤寒多价染色平板凝集试验抗原(以下简称为商品染色抗原)的检测结果及其与D群沙门氏菌抗体ELISA试剂盒检测结果的一致性。结果表明:商品染色抗原、自制染色抗原、自制凝集抗原和D群沙门氏菌抗体ELISA试剂盒检测的阳性率分别为50%、78%、40%和72%。商品染色抗原Cohen’s kappa系数仅为0.08,提示一致性较差;自制染色抗原和自制凝集抗原的Cohen’s kappa系数为分别为0.363和0.375,一致性一般。自制染色抗原检测结果的阳性符合率最高,可达87.50%;而自制凝集抗原的阴性符合率更高,可达96.43%。说明试验成功制备了鸡白痢沙门氏菌染色抗原和凝集抗原;与商品染色抗原相比,自制抗原与D群沙门氏菌抗体ELISA试剂盒的检测结果一致性更高。 In order to evaluate the effectiveness of self-made antigens for detection of Salmonella pullorum in chickens,in the experiment,from the results of screening by specific agglutination test and non-specific agglutination test,one clinical isolate of Salmonella pullorum SP-41 was randomly selected and mixed with the standard strain of Salmonella pullorum CVCC1792 in a ratio of 1∶1.Two methods,plate agglutination test and microagglutination test,were used to prepare staining antigen and microagglutination antigen,respectively;using 100 chicken serum samples as test subjects,the detection results of homemade antigen,Salmonella pullorum and Salmonella typhimurium multivalent staining plate agglutination test antigen(hereinafter referred to as commercial staining antigen),and their consistency with the detection results of group D Salmonella antibody ELISA kit were compared.The detection results showed that the positive rates of commercial staining antigen,homemade staining antigen,homemade agglutination antigen and group D Salmonella antibody ELISA kit were 50%,78%,40%and 72%,respectively.The Cohen's kappa coefficient of the commercial staining antigen was only 0.08,suggesting poor consistency;the Cohen's kappa coefficients of the homemade staining and agglutination antigens were 0.363 and 0.375,respectively,which showed average consistency.The positive compliance rate of homemade staining antigen was the highest,reaching 87.50%;while the negative compliance rate of homemade agglutination antigen was higher,reaching 96.43%.The results indicated that the test successfully prepared Salmonella pullorum staining antigen and agglutination antigen;compared with the commercial staining antigen,the homemade antigen had a higher consistency with the group D Salmonella antibody ELISA kit.
