辣蓼黄酮缓解猪伪狂犬病病毒诱导的小鼠炎症及相关信号通路研究

Study on the Ethyl Acetate Fraction of Flavonoids from Polygonum hydropiper L.Alleviates PRV-induced Inflammation and Related Signalling Pathways in Mice

摘要 为研究辣蓼黄酮乙酸乙酯部分(ethyl acetate of flavonoids from Polygonum hydropiper L,FEA)对猪伪狂犬病病毒(Pseudorabies virus,PRV)感染小鼠诱导的炎症反应及炎症相关信号通路的调节作用,以50~200 mg/kg体重的FEA处理PRV感染的小鼠炎症模型后,检测各组小鼠免疫器官指数变化,脾脏匀浆液中氧化应激相关因子、炎症相关因子的水平或表达量,以及NF-κB等炎症相关信号通路的活化情况。结果显示,50和200 mg/kg体重的FEA能降低PRV感染的小鼠脾脏指数(P<0.05);50~200 mg/kg体重FEA能不同程度的降低PRV感染的小鼠脾脏细胞中ROS、NO水平,IL-1β、IL-6、MCP-1和TNF-α等细胞因子的含量及mRNA表达,以及XOD、MPO、iNOS的酶活性(P<0.05),升高IL-10的水平和GSH的酶活性(P<0.05)。此外,FEA降低了PRV感染的小鼠脾脏细胞中p-p65/p65、p-JNK/JNK和p-p38/p38等炎症相关信号通路蛋白的磷酸化比值(P<0.05)。结果表明,FEA能减缓PRV诱导的小鼠氧化应激状态和炎症反应,发挥体内抗炎作用,其可能是通过抑制NF-κB和MAPK等炎症相关信号通路的活化发挥作用。 In order to investigate the regulatory effects of ethyl acetate fraction of flavonoids from Polygonum hydropiper L.(FEA)on inflammation and inflammatory signaling pathways induced by pseudorabies virus(PRV)infection in mice,PRV-infected mice inflammatory model was treated with FEA of 50-200 mg/kg body weight,the changes of immune organ indexes,the levels or expressions of oxidative stress-related factors,inflammation-related enzymes and factors in spleen homogenate,as well as the activation of NF-κB and other inflammation-related signaling pathways were detected.The results showed that FEA of 50 and 200 mg/kg body weight could reduce the spleen index of PRV-infected mice(P<0.05);50-200 mg/kg body weight FEA can lower the levels of ROS and NO,the contents or mRNA expressions of IL-1β,IL-6,MCP-1,TNF-αand other cytokines in spleen cells of PRV-infected mice,as well as the enzyme activities of XOD,MPO,iNOS(P<0.05),while the level of IL-10 and enzyme activities of GSH were increased(P<0.05).In addition,FEA decreased the phosphorylation ratios of inflammatory signaling pathway proteins such as p-P65/p65,p-JNK/JNK,and p-P38/p38 in PRV-infected mouse spleen cells(P<0.05).The results showed that FEA can relieve the oxidative stress and inflammatory response induced by PRV in mice,and play an anti-inflammatory role in vivo,possibly by inhibiting the activation of inflammation-related signaling pathways such as NF-κB and MAPK.
