摘要
目的研究赤地利总黄酮对肝肾代谢的安全性及其指导用量。方法采用灌胃法连续给药15 d,摘取肝脏和肾脏称重并计算脏器系数,检验血清丙氨酸转氨酶(alanine aminotransferase,ALT)、天冬氨酸转氨酶(aspartate transaminase,AST)、血肌酐(serum screatinine,Scr)、血尿素氮(blood urea nitrogen,BUN)水平,并进行肝肾组织病理切片。结果小鼠的肝脏系数出现剂量双向性、脾脏系数发生浓度负效应;ALT、AST与对照组差异不显著,Scr与对照组间差异有统计学意义,但其浓度间差异无统计学意义,低浓度组BUN值与对照组差异显著且处理组的BUN值存在浓度正效应(P<0.05),但低于对照组,肝脏组织气球样变呈浓度正效应,肾脏组织无明显变化。结论赤地利总黄酮,口服剂量不能超过1 mg/kg。
Objective To study the safety and dosage of total flavonoids from Polygonum chinense L.on liver and kidney.Methods The drug was administered by gavage for 15 days.Liver and kidney were extracted and weighed,and organ coefficients were calculated.Alanine aminotransferase(ALT),aspartate transaminase(AST),serum screatinine(Scr),blood urea nitrogen(BUN)levels were tested,and liver and kidney histological sections were performed.Results The liver coefficient of mice showed dose-bidirectional and spleen coefficient showed concentration negative effect;there was no significant difference between ALT,AST and control group,and there was statistical difference between Scr and control group,but there was no statistical difference between concentration.The BUN value of the low concentration group was significantly different from that of the control group,and the BUN value of the treatment group had a positive concentration effect(P<0.05),but it was lower than that of the control group.The balloon change of liver tissue showed a positive concentration effect,and the kidney tissue had no significant change.Conclusion When the total flavonoids from Polygonum chinense L.are used in health food,the oral dose should not exceed 1 mg/kg.
