大肠埃希氏菌疫苗生产用菌种种子批的建立与鉴定

摘要 为了研制仔猪大肠杆菌病三价(K88+K99+987P)灭活疫苗,以大肠埃希氏菌生产菌种C83549(k88)第2代和C83644(k99)、C83710(987p)第3代基础种子作为原始种子,建立了用于仔猪大肠杆菌病疫苗生产的大肠埃希氏菌基础种子批,分别对种子批的F1、F5、F10、F13代进行了全面鉴定。结果表明,各代种子的培养特性、生化特性均符合大肠埃希氏菌特征,抗原结构分别为C83549(O149∶K88)、C83644(O68∶K99)和C83710(O9∶987P);各代次菌株的菌毛抗原RIHA单位≥800,菌种免疫原性鉴定合格;各菌株经口服途径灌服1.5×10^(10)CFU/头,可引起仔猪100%发病。因此,3个菌株均可用作仔猪大肠杆菌病三价(K88+K99+987P)灭活疫苗生产的菌种。 In order to develop an inactivated vaccine against Escherichia coli in piglets,the second generation basic seeds of Escherichia coli strains C83549 and third generation basic seeds of C83644 and C83710 were used as the original seeds.A basic seed batch of Escherichia coli was established for the production of Escherichia coli vaccines in piglets,and the F1,F5,F10,and F13 generations were comprehensively identified.The cultivation and biochemical characteristics of each generation of seeds are consistent with the characteristics of Escherichia coli,with antigen structures of C83549(O149∶K88),C83644(O68∶K99),and C83710(O9∶987P),respectively;The RIHA unit of ciliated antigen in each generation of strains is≥800,and the immunogenicity of the strains is qualified;Each strain is orally administered 1.5 times×10^(10)CFU per head can cause 100%disease in piglets.Therefore,all three strains can be used as production strains for the trivalent inactivated vaccine against Escherichia coli(K88+K99+987P)in piglets for vaccine production.
