抑菌剂对白掌‘维美’组培苗生长的影响

摘要 [目的]探究不同抑菌剂对白掌‘维美’组培苗生长及污染率的影响,以期为白掌标准化生产提供技术支持。[方法]以植菌清、菌杀光、ZP抗菌素为抑菌剂,分别设置5种浓度梯度,以清水培养基为对照(CK)。在诱导、增殖分化和壮苗生根培养基中分别添加不同浓度的抑菌剂,比较不同抑菌剂处理下‘维美’诱导、增殖分化和壮苗生根阶段的抑菌效果,并探讨其对组培苗生长的影响。[结果]1.8 mL·L^(-1)植菌清处理下,‘维美’外植体诱导的污染率为10%,且组培苗的长势与CK差异不大;0.6 mL·L^(-1)菌杀光处理下,‘维美’的增殖系数与CK相同,均为4.2,且其污染率与畸变率均为0;10 mL·L^(-1) ZP抗菌素处理下,‘维美’生根培养的污染率为0,根长2.46 cm、根粗1.08 mm、生根数2.2,生根率达100%。[结论]适合‘维美’组培苗外植体诱导阶段、增殖分化阶段及壮苗生根阶段的抑菌剂分别为1.8 mL·L^(-1)植菌清、0.6 mL·L^(-1)菌杀光和10 mL·L^(-1) ZP抗菌素。在不同的培养阶段选择适宜的抑菌剂能够明显降低‘维美’组织培养的污染率。 [Purpose]To explore the effects of different bacteriostatic agents on the growth and contamination rate of tissue culture seedlings of Spathiphyllum floribundum‘Weimei’,further to provide technical support for the standardized production of S.floribundum.[Method]Five concentration gradients were set with Zhijunqing,Junshaguang,and ZP antibiotic as bacteriostatic agents,and medium added with water was used as control(CK).Different bacteriostatic agents were added to the medium during the induction,proliferation and differentiation,rooting stages,respectively.The effects of these bacteriostatic agents on bacteriostasis and the growth of‘Weimei’tissue culture seedlings during each stage were investigated.[Result]When treated with 1.8 mL·L^(-1) Zhijunqing,the contamination rate induced by‘Weimei’explants was 10%.There was no significant difference in the growth of tissue culture seedlings with CK.When treated with 0.6 mL·L^(-1) Junshaguang and CK,the proliferation coefficients of‘Weimei’were 4.2,and the contamination rate and distortion rate of 0.6 mL·L^(-1) Junshaguang were 0.When treated with 10 mL·L^(-1) ZP antibiotics,the contamination rate of‘Weimei’rooting seedlings was 0,the root length was 2.46 cm,the root diameter was 1.08 mm,the root number was 2.2,and the rooting rate was 100%.[Conclusion]The bacteriostatic agents suitable for‘Weimei’tissue culture seedlings during the stages of induction,proliferation,differentiation,and rooting were 1.8 mL·L^(-1) Zhijunqing,0.6 mL·L^(-1) Junshaguang,and 10 mL·L^(-1) ZP antibiotics,respectively.Selecting appropriate bacteriostatic agents at different culture stages can significantly reduce the contamination rate of‘Weimei’tissue culture.
