摘要 通过对胡萝卜新透心红 DaucuscarotaCV.XinTouxinhong 细胞悬浮系单位鲜重细胞的茄红素含量、细胞总产量以及茄红素总产量的分析比较,研究了培养基中不同NAA浓度对胡萝卜细胞悬浮系茄红素代谢的影响.结果表明,NAA具有增强细胞茄红素代谢水平、提高单位鲜重细胞中茄红素的含量的作用,但对细胞的生长具有抑制作用.从茄红素单产和总产量综合评价,培养基中加入3.0mg·L-1NAA效果最佳. Plant growth regulators have great impacts on the synthesis of secondary metabolites in plant cells in vitro. In order to study the effects of α naphthalene acid (NAA ) on lycopene synthesis in carrot suspension cell system, (Daucus carota CV.Xin Touxinhong) stable cell suspension system produced and contained in the medium of MS, containing vitamine in B5 medium, 2 mg·L-1 2,4D , 0.1 mg·L-1 KT and 30 g·L-1 sucrose was employed as the material, the favorite medium of MG5 selected from the media namely MS, White, B5, M9 , LS, ER and MG5 was used as basic medium. The concentration treatments of NAA in basic medium were(CK),0.5,1.0,3.0,5.0 and 10.0 mg·L-1. After cultured in cell growth medium of MS, containing vitamine in B5 medium, 0.1 mg·L-1 2,4D and 30 g·L-1 sucrose for two weeks, the carrot suspension cells collected with centrifugation were transferred into treatment media and cultured at 25℃ with 100~120 rpm oscillation in the dark. Two weeks later, total fresh / dry weights, lycopene content and total lycopene content of every treatment suspension cell system were measured. The results showed that the lycopene content of unit fresh weight cells in the medium with 3.0 mg·L-1 NAA was significantly higher than the other treatments including CK, the concentrations of NAA more than 5.0 mg·L-1 or less than 1.0 mg·L-1 inhibited the lycopene synthesis in the carrot suspension cells. The total content of lycopene in carrot cell suspension system was also the highest at the 3.0 mg·L-1 NAA concentration, other treatments were all lower than CK. Otherwise, the total weight of cells in the media with NAA were all lower than the CK. In conclusion, addition of NAA in the media promoted the lycopene metabolism and upgraded the lycopene content of unit fresh weight cells, and repressed the cell growth compared with control. The optimum concentration of NAA in the medium for lycopene synthesis in carrot cells was 3.0 mg·L-1.
