骨组织工程材料生长因子复合物与成骨细胞相容性研究 被引量:2

摘要 目的 :研究复合有生长因子的骨组织工程材料与成骨细胞的相容性 ,以及所复合的生长因子对成骨细胞增殖、分化的作用。方法:自新生大鼠颅盖骨分离培养成骨细胞接种于复合有BMP_2、IGF_1等生长因子的骨组织工程材料上。培养72h后计算材料上所附着的细胞数以及细胞内碱性磷酸酶的活性。结果:材料上所复合的BMP_2可明显增加成骨细胞内碱性磷酸酶的活性,但对细胞的增殖无影响。多聚赖氨酸可促进细胞对材料的黏附 ,IGF_1对成骨细胞的增殖促进作用明显。结论:复合有生长因子的骨组织工程材料与成骨细胞的相容性良好 ,材料上所载的生长因子对所黏附的成骨细胞增殖。 Objective:To study the compatibility of osteoblast with the comp ound of bone tissue_engineering material and growth factors,and the effect of growth factor on proliferation and differentiation of osˉteoblast.Methods:Osteoblast isolated and cultured from newborn rat calvaris was seeded on the tisˉsue_engineering materials compounded with BMP_2and IGF_1.The number of the cell adhered on the materials and the activity of ALP were calculated72hours after the culture.Results:BMP_2could increase the activity of ALP obviously.Polylysine could enhance the adhesion of cell onto the artificial bone material.IGF_1increased the proliferation of osteoblast markedly.Conclusion:There was a good compatibility between osteoblast and bone tissue_enginering material compounded with growth factors.BMP_2and IGF_1could increase the proliferˉation and differentiation of osteoblast adhered on materials.
