摘要
目的 :寻找一种简便、快捷、灵敏的观察骨折愈合过程中胶原类型、分布动态变化的方法。方法 :在健康中国绵羊左跖骨中段制造一个 1cm长的骨缺损。分别在术后 1、 3、 6个月取材 ,苦味酸天狼星红染色 ,偏振光显微镜观察。结果 :术后 1个月 ,骨折局部Ⅲ型胶原纤维占大多数 ;术后 3个月 ,骨折处Ⅰ型胶原纤维明显增加 ,但排列尚未有序 ;术后 6个月 ,Ⅰ型胶原纤维增粗、增多 ,呈斜行螺旋状交叉排列。结论 :苦味酸天狼星红偏振光不仅能区分Ⅰ、Ⅲ型胶原的类型还能清晰地显示Ⅰ、Ⅲ型胶原的形态、分布和比例关系 ,具有操作简单、省时、花费少 ,而且特异性强、灵敏度高。
Objective:To find a convenient,swif and sensitive method for observation of the developing change of collagen fibers' type and distribution during fracture healing.Method:A 1cm bone defect was made in left metatarsus of the sheep,then euthanasia was and bone specimens were harvested at 1st,3rd and 6th month.The specimens were stained by picrosirius and observed by polarimicroscope.Result:Collagen Ⅲ occupied mostly the fracture site 1 month post-operatively;collage Ⅰ increased evidently at the third month,but its array was still not regular;collagen Ⅰ was not only thicker and much more in original defect,but also took a gyrate arrangement.Conclusion:Picrosirius stain can clearly display the distribution,form and proportion of collagen Ⅰ and Ⅲ.This method is simple,convenient,cheap,specific and highly sensitive.
