维生素E治疗大鼠少弱精症的效果及作用机制的研究 被引量:3

摘要 目的:观察维生素E治疗少弱精症模型大鼠的效果,并探讨作用机制。方法:选择健康Sprague-Dawley大鼠28只,按随机数字表法分为四组:正常对照组、模型组、溶剂治疗组和维生素E治疗组,各7只。采用雷公藤多甙诱导少弱精症的模型大鼠,维生素E治疗组灌胃维生素E(150 mg/kg)连续5周,溶剂治疗组灌胃等体积的玉米油。比较各组大鼠精子的密度、活力、活率以及直线运动速度等,以及睾丸组织中HSP70-2蛋白的表达。结果:雷公藤多甙组大鼠精子的密度、活率、活力以及直线运动速度等指标低于正常组,差异有统计学意义(P<0.05);维生素E治疗组大鼠精子的密度、活率、活力以及直线运动速度高于溶剂治疗组,差异有统计学意义(P<0.05);雷公藤多苷组大鼠睾丸中HSP70-2蛋白的表达低于正常组,差异有统计学意义(P<0.05);维生素E治疗组HSP70-2蛋白的表达高于溶剂治疗组,差异有统计学意义(P<0.05)。结论:维生素E对少弱精症具有明显的治疗作用,可能是通过上调睾丸组织中HSP70-2蛋白的表达实现的。 Objective:To observe the effect of vitamin E in the treatment of oligozoospermia in rats,and to explore its action mechanism.Methods:28 healthy Sprague-Dawley rats were randomly divided into four groups:a normal control group,a model group,a solvent treatment group and a vitamin E treatment group,7 rats each.Tripterygium wilfordii polyglycosides were used to induce oligozoospermia in rats.The rats in the vitamin E treatment group were treated with vitamin E(150 mg/kg)by lavage for 5 weeks,and the rats in the solvent treatment group were treated with an equal volume of corn oil by lavage.The density,vitality,survival rate and linear motion velocity of rat sperms,and the expression of HSP70-2 protein in testis tissues in each group were compared.Results:The density,vitality,survival rate and linear motion velocity of the rat sperms in the Tripterygium wilfordii polyglycoside group were lower than those in the normal group,and the differences were statistically significant(P<0.05).The density,vitality,survival rate and linear motion velocity of the rat sperms in the vitamin E treatment group were higher than those in the solvent treatment group,and the differences were statistically significant(P<0.05).The expression of HSP70-2 protein in testis of rats in the Tripterygium wilfordii polyglycoside group was lower than that in the normal group,and the difference was statistically significant(P<0.05).The expression of HSP70-2 protein in the vitamin E treatment group was higher than that in the solvent treatment group,and the difference was statistically significant(P<0.05).Conclusion:Vitamin E has a significant therapeutic effect on oligozoospermia,which may be possibly by up-regulating the expression of HSP70-2 protein in testis tissues.
