肿节风抗炎镇痛及对盆腔炎模型大鼠的影响 被引量:7

Anti-Inflammatory and Analgesic Effect of SARCANDRAE HERBA and Its Influence on Rat Model of Pelvic Inflammation

摘要 目的:研究肿节风的抗炎镇痛作用及对盆腔炎模型大鼠的影响,并初步探讨其机制。方法:采用3种炎症模型和3种疼痛模型研究肿节风的抗炎镇痛作用;采用机械损伤和混合细菌感染双重损伤方法建立大鼠盆腔炎模型,造模成功后,将大鼠随机分为模型对照组、肿节风1.75、3.5、4.5、7、9 g/kg组及妇科千金片0.672 g/kg组,另设正常对照组,各组给予相应药物或生理盐水连续14 d。末次给药后12 h后,取大鼠双侧子宫,HE染色法观察子宫组织形态学改变,ELISA法测定大鼠血清中前列腺素(PGE_(2))、组胺和成纤维细胞生长因子(FGF-2)的含量。结果:肿节风4.5、9 g/kg组能明显抑制小鼠的耳肿胀程度,抑制小鼠腹腔毛细血管通透性的增加,减轻棉球致肉芽组织的质量(P<0.05);延长醋酸致小鼠疼痛反应的潜伏时间并减少其扭体次数,减少甲醛致痛引起的舔足次数(P<0.05或P<0.01);肿节风3.5、7 g/kg组能显著改善盆腔炎模型大鼠子宫组织病理损伤,降低大鼠血清中PGE_(2)、组胺及FGF-2的含量(P<0.05或P<0.01)。结论:肿节风有显著的抗炎作用及一定的镇痛作用,对大鼠盆腔炎模型有明确的治疗作用,其作用以抗炎镇痛为基础,可能与抑制血清中PGE_(2)和组胺的释放,降低FGF-2的含量有关。 Objective:To study the anti-inflammatory and analgesic effect of SARCANDRAE HERBA and its influence on rat model of pelvic inflammation,primarily exploring its possible mechanism.Methods:Three inflammatory models and three pain models were used to study the anti-inflammatory and analgesic effect of SARCANDRAE HERBA.The rat model of pelvic inflammation was induced by mechanical injury and mixed bacterial infection.After successful modeling,the rats were randomly divided into a blank control group,a model control group,four SARCANDRAE HERBA(1.75,3.5,4.5,7,and 9 g/kg)groups,and a Fukeqianjin(妇科千金)tablet(0.672 g/kg)group,and each group was given corresponding drugs for 14 d.The bilateral uteruses of the rat were collected 12 h after the last administration,and the histomorphological changes in uterus were observed by hematoxylin-eosin(HE)staining.The content of prostaglandin E2(PGE_(2)),histamine,and fibroblast growth factor-2(FGF-2)in the serum were determined by enzyme-linked immunosorbent assay(ELISA).Results:The 4.5 and 9 g/kg SARCANDRAE HERBA significantly reduced the degree of ear swelling,inhibited the increase of celiac capillary permeability in rats,and reduced the weight of granulation tissue caused by cotton balls(P<0.05).In the 4.5 and 9 g/kg SARCANDRAE HERBA groups,the latency of pain response induced by acetic acid was prolonged,and the number of writhing and foot licks induced by formaldehyde in rats was reduced(P<0.05 or P<0.01).The 3.5 and 7 g/kg SARCANDRAE HERBA significantly reduced the pathological injury of the uterine tissue and reduced the content of PGE_(2),histamine,and FGF-2 in serum of rats with pelvic inflammation(P<0.05 or P<0.01).Conclusion:SARCANDRAE HERBA has significant anti-inflammatory and analgesic effects,and has obvious therapeutic effect on the rat model of pelvic inflammation.Its effect is based on anti-inflammation and analgesia,and the mechanism may be related to inhibiting the release of PGE_(2)and histamine in serum and down-regulating the level of FGF-2.
