天山堇菜提取物对环磷酰胺诱导免疫低下小鼠免疫功能的影响 被引量:1

Effects of Viola Tianshanica Maxim Extract On Immune Function of Immunosuppressed Mice Induced by Cyclophosphamide

摘要 目的:探讨天山堇菜提取物对环磷酰胺所致免疫低下小鼠免疫功能的影响。方法:将ICR小鼠随机分为正常对照组、模型对照组、贞芪扶正颗粒3.9 g/kg组和天山堇菜提取物0.08、0.16、0.32 g/kg组。除正常对照组外,其余各组小鼠腹腔注射环磷酰胺制备免疫低下模型,连续给药10 d后测定小鼠脾脏指数、胸腺指数;碳粒廓清法检测小鼠巨噬细胞吞噬功能;MTS法检测小鼠脾脏T、B淋巴细胞增殖能力;ELISA法测定小鼠血清中肿瘤坏死因子-α(TNF-α)、γ-干扰素(IFN-γ)和免疫球蛋白M(IgM)含量。结果:与正常对照组相比,模型对照组小鼠体质量、脾脏指数、胸腺指数、碳廓清指数、吞噬指数、脾脏淋巴细胞增殖能力及血清TNF-α、IFN-γ和IgM含量均明显降低(P<0.05或P<0.01),表明免疫抑制小鼠模型建立成功;与模型对照组相比,天山堇菜提取物0.08、0.16、0.32 g/kg组均能明显促进免疫抑制小鼠血清中IFN-γ的分泌(P<0.05或P<0.01);天山堇菜提取物0.16、0.32 g/kg组均能明显促进免疫抑制小鼠血清中IgM的分泌(P<0.05),促进脾脏B淋巴细胞增殖(P<0.01);天山堇菜提取物0.32 g/kg组能明显升高免疫抑制小鼠脾脏指数,升高碳廓清指数和吞噬指数,促进脾脏T淋巴细胞增殖,明显增加血清中TNF-α的含量(P<0.05)。结论:天山堇菜提取物对免疫功能低下小鼠的免疫功能有增强作用。 Objective:To investigate the effects of Viola tianshanica Maxim extract(VTE)on the immune function of immunosuppressed mice induced by cyclophosphamide.Methods:ICR mice were randomly divided into a normal control group,a model control group,a Zhenqi Fuzheng(贞芪扶正)Granules group(3.9 g/kg),and low-,medium-,and high-dose VTE groups(0.08,0.16,and 0.32 g/kg).The immunosuppression model was induced by intraperitoneal injection of cyclophosphamide in mice except for those in the normal control group.After 10 days of drug intervention,spleen index and thymus index of immunosuppressed mice were measured.The phagocytic function of macrophages was determined by carbon clearance test.MTS assay was used to detect the proliferation of splenic T and B lymphocytes.The serum levels of tumor necrosis factor(TNF)-α,interferon(IFN)-γ,and immunoglobulin M(IgM)in mice were observed by enzyme-linked immunosorbent assay(ELISA).Results:Compared with the normal control group,the model control group showed reduced body mass,spleen index,thymus index,clearance index K,and phagocytic index,blunted proliferation of splenic T and B lymphocytes,and decreased serum levels of TNF-α,IFN-γ,and IgM(P<0.05 or P<0.01),which suggested the immunosuppression model was properly induced.Compared with the model control group,the VTE groups showed increased serum levels of IFN-γ(P<0.05 or P<0.01).The medium-and high-dose VTE increased the secretion of IgM(P<0.05)and promoted the proliferation of B lymphocytes(P<0.01).In addition,the high-dose VTE remarkably enhanced the spleen index,the clearance index K,and phagocytic index,promoted the proliferation of T lymphocytes,and increased the serum levels of TNF-α(P<0.05).Conclusion:VTE can enhance the immune function of immunosuppressed mice induced by cyclophosphamide.
