古代医籍中治疗筋病内服药规律的数据挖掘研究

Data Mining Research on the Rule of Internal Drugs in the Treatment of Jin Disease in Ancient TCM Books

目的:探讨古代医籍中治疗筋病内服方剂的用药与组方规律,以期对筋病的临床治疗及新药研发提供参考。方法:收集整理《中华医典单机版V5.0》中治疗筋病共129首内服方剂,涉及186味中药,录入中医传承辅助系统(V2.5),采用改进的互信息法、关联规则、核心算法、熵层次聚类,统计单药、对药、角药出现频次,并分析关联规则、提取核心组合、发现新方,进行数据挖掘研究。结果:最常见单药是当归(78次,占41.94%);最常见对药是当归-肉桂(39次,占30.23%);最常见角药是没药-乳香-当归(21次,占16.28%)、赤芍-川芎-当归(21次,占16.28%);关联度最高的药物组合是乳香+当归→没药(置信度为1.0000);得到24组3至4味药的核心组合,核心药物为:当归、肉桂、没药、川芎、赤芍、乳香等;得到内服治疗筋病12个新候选处方。结论:提示中药内服治疗筋病以及新药研发时,单药、对药、角药优先考虑当归、肉桂、没药、乳香;配伍优先考虑乳香、当归配伍没药;核心方剂优先考虑木香、沉香、藿香等;新候选处方优先考虑木香、沉香、藿香、独活等。

Objective:To investigate the prescription rules of internal herbal medicines in the treatment of Jin disease in ancient TCM books,so as to provide a reference for clinical treatment for Jin disease,and research and development of new drugs.Methods:A total of 129 traditional Chinese prescriptions containing 186 herbs from Zhonghuayidian Standalone Version V5.0 were collected and input into TCM Inheritance Auxiliary Plat form Version 2.5.Methods of improved mutual information entropy clustering,the association rules,the complex systems entropy clustering and the unsupervised entropy hierarchical clustering were used to calculate the using frequency of single drug,drug pairs and three-drug combination,then the association rules,the evolution and extraction of combination of the core drugs,and new prescriptions were analyzed and the data mining was performed.Results:The most commonly used herbal medicine was Angelica(78,41.94%),and the most common drug pair was Angelica-Cassia(39,30.23%),the most common three-drug combinations were Myrrh-Frankincense-Angelica(21,16.28%)and Red Paeonia-Rhizoma Chuanxiong-Angelica(21,16.28%).The drug combination with high correlation was Frankincense+Angelica→Myrrh(confidence 1.0000).Totally 24 groups of three-drug or four-drug combinations were extracted,in which the core drugs were Angelica,Cassia,Myrrh,Rhizoma Chuanxiong,Red Paeonia and Frankincense.And 12 new prescriptions were screened.Conclusion:In research and development of new internal drugs in the treatment of Jin disease,Angelica,Cassia,Myrrh and Frankincense should be top-priority.In prescription rules,Frankincense,Angelica combined with Myrrh should be top-priority.Radix Aucklandiae,Chinese Eaglewood Wood,Pogostemon Cablin,etc should be the top-priority core prescriptions.And Radix Aucklandiae、Chinese Eaglewood Wood、Pogostemon Cablin、Radix Angelicae Pubescentis etc.were considered the candidate prescription for the treatment of Jin disease.
