白术水提物对自然衰老小鼠的作用研究

Effect of Baizhu(白术)Water Extract on Natural Aging in Mice

目的:观察白术水提物对自然衰老小鼠的影响。方法:采用自然衰老小鼠,灌胃给予白术水提物1、2、4 g/kg,连续4 w。期间检测小鼠抓力、自主活动次数、腹温和避暗穿梭次数;测定全血中白细胞(WBC)、红细胞(RBC)、血红蛋白(HGB)、淋巴细胞(LYMPH)、血小板(PLT)和网织红细胞(RET)数量;流式细胞仪检测外周血T细胞、B细胞、NK细胞、Th细胞、Tc细胞、初始性Th细胞、记忆性Th细胞、效应性Th细胞、初始性Tc细胞、记忆性Tc细胞、效应性Tc细胞水平。结果:与正常对照组相比,模型对照组小鼠衰老表现评分、自主活动次数和避暗穿梭次数明显升高(P<0.05或P<0.01),腹温和抓力显著降低(P<0.01);全血WBC和LYMPH数量显著降低(P<0.01),PLT和RET数量明显升高(P<0.05或P<0.01);外周血T细胞、Th细胞、记忆性Th细胞和初始性Tc细胞水平明显降低(P<0.05或P<0.01),Tc细胞和效应性Th细胞显著升高(P<0.01)。与模型对照组相比,白术水提物3个剂量组在一定程度上可明显降低自然衰老小鼠衰老表现评分、自主活动次数和避暗穿梭次数(P<0.05或P<0.01),升高腹温和抓力(P<0.05或P<0.01);升高全血WBC、RBC、HGB、LYMPH和RET数量(P<0.05或P<0.01),降低PLT数量(P<0.05或P<0.01);升高外周血T细胞及初始性Th细胞水平(P<0.05或P<0.01)。结论:白术水提物能改善自然衰老小鼠免疫低下、造血不足和记忆衰减等衰老相关功能减退。

Objective:To observe the effect of Baizhu(白术)water extract on natural aging in mice.Methods:The natural aging mice were gavaged with 1,2,and 4 g/kg Baizhu(白术)water extract for four successive weeks.During the medication,the grasping force,number of autonomous activities,abdominal temperature,and number of dark avoidance shuttle were measured.The white blood cell(WBC),red blood cell(RBC),hemoglobin(HGB),lymphocyte(LYMPH),platelet count(PLT),and reticulocyte count(RET)in the whole blood were determined.The T cells,B cells,NK cells,Th cells,Tc cells,primary Th cells,Th memory cells,Th effector cells,primary Tc cells,Tc memory cells,and Tc effector cells in the peripheral blood were determined by flow cytometry.Results:Compared with the normal control group,the model group exhibited significantly elevated score of aging performance,number of autonomous activities,and number of dark avoidance shuttle(P<0.05 or P<0.01),decreased abdominal temperature and grasping force(P<0.01),lowered WBC and LYMPH(P<0.01)as well as T cells,Th cells,Th memory cells,and primary Tc cells in the peripheral blood(P<0.05 or P<0.01),raised PLT and RET(P<0.05 or P<0.01)as well as Tc cells and Th effector cells(P<0.01).Compared with the model group,Baizhu(白术)water extract at the three doses significantly reduced the aging performance score,the number of autonomous activities,and the number of dark avoidance shuttle in natural aging mice to a certain extent(P<0.05 or P<0.01),increased the abdominal temperature and grasping force(P<0.05 or P<0.01),WBC,RBC,HGB,LYMPH,and RET in the whole blood(P<0.05 or P<0.01),and T cells and primary Th cells in the peripheral blood(P<0.05 or P<0.01),and decreased the number of PLT(P<0.05 or P<0.01).Conclusion:Baizhu(白术)water extract improves the aging related function decline like hypoimmunity,anematopoiesis,and memory loss.
