鸡矢藤提取物对幼龄厌食症模型大鼠的影响

Effect of Jishiteng(鸡矢藤)Extract on Anorexia in Young Model Rats

目的:研究鸡矢藤提取物对幼龄厌食症模型大鼠的治疗作用及可能的作用机制。方法:采用特殊饲料饲养,构建幼龄厌食症模型大鼠,观察鸡矢藤提取物10、20、30 g/kg对动物体质量、体质量增长、摄食量、胃肠动力的影响;Elisa法检测血清中胃肠激素胃动素(MTL)、胃泌素(GAS),脑肠肽饥饿素(Ghrelin)、β-内咖肽(β-EP)、生长抑素(SS)含量;免疫组织化学法检查结肠组织Cajal间质细胞(ICC)C-kit蛋白的表达。结果:与正常对照组比较,模型对照组动物体质量、体质量增长、摄食量、胃肠动力、MTL、GAS、Ghrelin、β-EP含量明显降低,SS明显升高,结肠C-kit蛋白表达降低(P<0.01)。鸡矢藤提取物30 g/kg可使动物体质量、体质量增长、摄食量、胃肠动力增加、MTL、GAS、Ghrelin、β-EP含量增加,SS含量显著降低,结肠C-kit蛋白表达增加(P<0.05或者P<0.01)。结论:鸡矢藤提取物30 g/kg对幼龄厌食症模型大鼠有明显治疗作用,其作用机制可能与降低脑肠肽SS含量、促进Ghrelin、β-EP分泌,MTL、GAS胃肠激素释放,调节ICC细胞C-kit蛋白的表达,改善胃肠动力有关。

Objective:To study the therapeutic effect of Jishiteng(鸡矢藤)extract on anorexia in young model rats and the possible mechanism.Methods:The young rat model of anorexia was established by exposing them to special fodder,and the effects of Jishiteng(鸡矢藤)extract at different doses(30,20,and 10 g/kg)on body weight,weight gain,food intake,and gastrointestinal dynamics were observed.The levels of serum gastrointestinal hormones motilin(MTL),gastrin(GAS),Ghrelin,β-endorphin(β-EP),and somatostatin(SS)were measured by Elisa,and the C-kit protein expression in the interstitial cells of Cajal(ICCs)of colon tissue was examined by immunohistochemistry.Results:Compared with the normal control group,the model group displayed significantly reduced body weight,weight gain,food intake,gastrointestinal dynamics,MTL,GAS,Ghrelin,andβ-EP,increased SS,and down-regulated C-kit protein expression(P<0.01).Jishiteng(鸡矢藤)extract at 30 g/kg elevated the body weight,weight gain,food intake,gastrointestinal dynamics,MTL,GAS,Ghrelin,andβ-EP,lowered SS,and up-regulated C-kit protein expression(P<0.05 or P<0.01).Conclusion:Jishiteng(鸡矢藤)extract at 30 g/kg has a significant therapeutic effect on anorexia in young model rats,which may be related to its reduction of SS,induction of Ghrelin andβ-EP secretion and MTL and GAS release,regulation of C-kit protein expression in ICCs,and improvement of gastrointestinal dynamics.
