滇白珠活性部位对腹泻大鼠结肠组织病理学和水通道蛋白3表达的影响

Effects of Active Fraction of Dianbaizhu(滇白珠)on Colonic Histopathological Change and Aquaporin 3 Expression in Diarrhea Rats

目的:研究滇白珠活性部位对腹泻大鼠结肠组织病理学和水通道蛋白(aquaporin 3,AQP3)表达的影响。方法:60只雄性大鼠随机分为正常对照组、模型对照组、洛哌丁胺6 mg/kg组和滇白珠活性部位40、80、160 mg/kg组,每组10只。各组动物每天灌胃给药1次,连续12 d,正常对照组、模型对照组给予同容积溶媒。第3 d起,除正常对照组外,其余5组在给药1 h后,连续10 d按10 mL/kg灌胃给予3.33 g/kg番泻叶提取物造腹泻模型,正常对照组灌胃给予等容量的溶媒。末次给予番泻叶提取物后,观察滇白珠活性部位对大鼠稀便率、稀便等级和腹泻系数的影响。采用苏木精-伊红(HE)染色观察滇白珠活性部位对腹泻大鼠结肠组织病理学改变的影响。采用Western blot和RT-PCR分析滇白珠活性部位对腹泻大鼠结肠组织AQP3蛋白和mRNA表达的影响。结果:滇白珠活性部位40、80、160 mg/kg能显著降低腹泻大鼠稀便率、稀便等级和腹泻系数,抑制结肠腺体内黏液的渗出,减轻肠黏膜及间质损伤(P<0.05或P<0.01)。Western blot和RT-PCR结果显示,滇白珠活性部位160 mg/kg能显著上调腹泻大鼠结肠组织AQP3的蛋白和基因的表达(P<0.01)。结论:滇白珠活性部位具有止泻作用,能有效保护结肠黏膜和间质细胞,上调结肠组织AQP3蛋白和基因的表达。

Objective:To study the effects of the active fraction of Dianbaizhu(滇白珠)on colonic histopathological change and aquaporin 3(AQP3)expression in diarrhea rats.Methods:Sixty male rats were randomly divided into the normal control group,model group,6 mg/kg loperamide group,and 40,80,and 160 mg/kg Dianbaizhu groups,with 10 in each group.The rats in medication groups were treated with intragastric administration of the corresponding drugs,once a day,for 12 consecutive days,while those in normal control group and model group with the same volume of solvent.From the 3 rd day,rats in all groups except for the normal control group were also gavaged with 3.33 g/kg Fanxieye(番泻叶)extract at 10 mL/kg for 10 consecutive days 1 h after administration,whereas those in the normal control group with the same volume of solvent.After the last administration of Fanxieye(番泻叶)extract,the effects of active fraction of Dianbaizhu(滇白珠)on the rate and grade of loose stools and the diarrhea coefficient of rats were observed.The pathological changes in rat colon after treatment with active fraction of Dianbaizhu(滇白珠)were observed by hematoxylin-eosin(HE)staining.The effects of active fraction of Dianbaizhu(滇白珠)on AQP3 protein and mRNA expression in colon tissues of rats with diarrhea were detected by Western blotting and RT-PCR.Results:The active fraction of Dianbaizhu(滇白珠)at 40,80,and 160 mg/kg significantly reduced the rate and grade of loose stools and the diarrhea coefficient,inhibited the exudation of mucous in the colonic glands,and alleviated the intestinal mucosa and interstitial injuries.Western blot and RT-PCR results showed that the AQP3 protein and gene expression levels in colon tissue of diarrheal rats were significantly up-regulated by the active fraction of Dianbaizhu(滇白珠)at 160 mg/kg.Conclusion:The active fraction of Dianbaizhu(滇白珠)is effective in inhibiting diarrhea,protecting colonic mucosa and mesenchymal cells,and up-regulating the AQP3 protein and gene expression in colonic tissues.
