杜仲补天素胶囊对雷公藤多苷所致雄性大鼠生殖障碍干预作用的实验研究 被引量:2

Experimental Study on the Intervention Effect of Duzhong Butiansu Capsule on Reproductive Disorders Induced by Tripterygium Glycosides in Male Rats

摘要 目的:研究杜仲补天素胶囊对雷公藤多苷所致雄性大鼠生殖障碍的干预作用并初步探讨其作用机制。方法:雄性SD大鼠随机分为空白对照组、溶剂对照组、模型组、生精胶囊560 mg/kg组、杜仲补天素485 mg/kg、970 mg/kg组。以雷公藤多苷(40 mg/kg)造模成功后,持续给药干预四周,雌雄合笼后,观察雌鼠受孕情况与产仔数,计算雄鼠脏器系数、测定各组性激素水平和氧化应激指标、观察睾丸组织形态变化、检测睾丸凋亡相关蛋白与雌激素受体蛋白表达的变化。结果:与正常组相比,模型组睾丸曲细精管空泡化,曲细精管内各级精细胞不同程度减少;睾丸系数、精子数量和精子活力均显著下降;受孕率与产仔数显著下降;雌二醇(E2)、睾酮(T)、卵泡雌激素(FSH)、促黄体生成素(LH)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)的含量或活力显著下降,丙二醛(MDA)的含量显著上升;睾丸组织中B淋巴细胞瘤-2蛋白(Bcl-2)表达显著下调,Bcl-2相关X蛋白(Bax)表达显著上调;睾丸中雌激素受体α和雌激素受体β(ERα、ERβ)蛋白的表达量显著上调。给药干预后与模型组相比,杜仲补天素胶囊组剂量依赖地使睾丸中曲细精管空泡化情况改善,各级精细胞再生;睾丸系数、精子数量和精子活力显著上升;受孕率与产仔数显著上升;E2、FSH、GSH-Px、SOD的含量或活力显著上升,MDA的含量显著下降;睾丸组织中Bax蛋白表达显著下调,Bcl-2蛋白表达显著上调;睾丸中ERα、ERβ蛋白的表达量显著下调。结论:杜仲补天素胶囊对雷公藤多苷所致雄性大鼠生殖障碍有一定的改善作用,可能是通过调控ERα、ERβ来调节氧化应激与凋亡水平实现的。 Objective:To investigate the intervention effect of Duzhong Butiansu Capsule on reproductive disorders in male rats induced by tripterygium glycosides and to explore its mechanism.Methods:Male Sprague-Dawley rats were randomly divided into the control group,the solvent control group,the model group,Shengjing capsule group(560 mg/kg)and Duzhong Butiansu groups(485 mg/kg,970 mg/kg).After successfully modeled by tripterygium glycosides(40 mg/kg),the rats were administered for four weeks.After the male and female rats were kept in one cage,the pregnancy and the litter size of the female rats were observed.The organ coefficient of male rats was calculated,the levels of sex hormones and Oxidative stress index in each group were determined.The morphology changes of testicular tissues,testicular apoptosis related proteins and estrogen receptor protein expression were observed.Results:Compared with the control group,the seminiferous tubules of the testis was vacuolated in the model group,the sperm cells in the seminiferous tubules were reduced to varying degrees,the testicular coefficient,sperm count and sperm motility were significantly decreased,the conception rate and litter size were significantly decreased.The contents or activities of E2,T,FSH,LH,GSH-Px and SOD were significantly decreased,and the content of MDA was significantly increased.The expression of Bax protein in the testicular tissue was significantly increased,and the expression of Bcl-2 protein was significantly decreased.The expressions of ERαand ERβin testicular were significantly increased.Compared with the model group,the vacuolization of the seminiferous tubules in the testis was improved in Duzhong Butiansu group,in dose dependant manner.The testicular coefficient,sperm count and sperm motility were significantly increased,the conception rate and litter size were significantly increased,levels of E2,T,FSH,GSH-Px and SOD were significantly increased,MDA content was significantly decreased.Bax protein expression in testicular tissue was significantly down regulated,while Bcl-2 protein expression level was significantly up regulated,the amounts of expressions of testicular ERα,ERβwere significantly reduced.Conclusion:Duzhong Butiansu Capsule can improve the reproductive disorder of male rats induced by tripterygium glycosides,it may be achieved by ERαand ERβregulating oxidative stress and apoptosis.
