摘要
目的:考察安息香不同剂量防治给药对永久性局灶性脑缺血(pMCAO)模型大鼠的影响,为阐明安息香脑保护的作用机制及量效关系提供参考。方法:采用改良线栓法制备pMCAO模型,参照mNSS和Zea Longa评分法,评价大鼠脑缺血各时间点的神经功能;测量给药30 min后大鼠体温,观察大鼠体温变化;TTC染色观察大鼠脑梗死率;ELISA法检测血清中VEGF、TEK、ANG-1、TGF-β1含量;HE染色观测缺血侧脑组织形态。结果:与假手术组比较,模型组大鼠体温、神经功能评分、脑梗死率均显著升高,模型组和溶剂模型组血清VEGF、TEK、ANG-1、TGF-β1浓度均显著降低,海马、神经元结构异常。与模型组比较,溶剂模型组各指标均无显著性差异;尼莫地平组大鼠神经功能评分、脑梗死率显著降低;安息香0.5 g/kg和1 g/kg组体温、神经功能评分、脑梗死率显著降低,VEGF、TEK、ANG-1、TGF-β1浓度显著升高,可显著改善皮层及海马CA1区损伤;安息香0.25 g/kg组体温、脑缺血48 h、72 h神经功能评分显著降低。结论:安息香通过活血、开窍醒神功效改善脑缺血损伤进而发挥脑保护作用。
Object:To investigate the effects of different doses of Benzoin on permanent focal cerebral ischemia(pMCAO)rats,and to provide a reference for dose-effect relationship and mechanism of Benzoin brain protection.Methods:The pMCAO model was established by modified suture method.The nerve functions of cerebral ischemia rats at different time were evaluated by mNSS and Zea Longa scoring method.The body temperature was measured 30 minutes after administration,and the difference was compared among the groups.Cerebral infarction rate was observed by TTC staining.The contents of VEGF,TEK,ANG-1 and TGF-β1 in serum were detected by ELISA.The morphology of ischemic brain tissue was observed by HE staining.Results:Compared with the sham operated group,the body temperature,neurological score and cerebral infarction rate were significantly increased in the model group.The serum levels of VEGF,TEK,Ang-1 and TGF-β1 in both solvent model group and model group were significantly decreased,and the structure of hippocampus and neurons were abnormal.Compared with the model group,no significant difference was observed in solvent model group,the neurologic function score and cerebral infarction rate in nimodipine group were significantly reduced.In 0.5 g/kg and 1 g/kg Benzoin groups,the body temperature,neurological score and cerebral infarction rate were significantly reduced,and the serum concentrations of VEGF,TEK,Ang-1 and TGF-β1 were significantly increased,which significantly improved the damage of cortex and hippocampal CA1 area.In 0.25 g/kg Benzoin group,the body temperature,neurological score at 48 h and 72 h of cerebral ischemia were significantly reduced.Conclusion:Benzoin can improve the cerebral ischemic injury by Huoxue and Kaiqiaoxingshen effect,then play a protective role in the brain.
