四磨汤对幼龄小鼠厌食症模型的影响 被引量:8

Therapeutic Effect and Mechanism of Simo Tang on the Anorexia Model in Young Mice

摘要 目的:研究四磨汤对幼龄小鼠厌食症的治疗作用及可能的作用机制。方法:采用特制饲料饲养的方法建立幼龄小鼠厌食症模型,观察不同剂量四磨汤(5 mL/kg、10mL/kg、20 mL/kg)对其体重增长、摄食量、胃肠动力的影响,HE染色观察胃肠道病理变化,ELISA法检测下丘脑和血清中脑肠肽胆囊收缩素(CCK-8)、生长抑素(SS)、β-内啡肽(β-EP),以及血清中胃泌素(GAS)、胃动素(MTL)的浓度变化,免疫组化检测胃窦黏膜中SS、GAS表达水平。结果:与空白对照组比较,模型对照组体重增长、摄食量、胃肠动力、血清中胃肠激素MTL、GAS含量显著降低,以及胃窦粘膜中GAS表达显著下调,下丘脑和血清中脑肠肽CCK-8、SS含量显著升高,胃窦粘膜中SS表达均显著上调,胃粘膜充血,胃、十二指肠表层上皮细胞变性坏死及松解脱落;与模型对照组比较,四磨汤20 mL/kg给药2周后,小鼠体重增长、摄食量、胃肠动力、血清中胃肠激素MTL、GAS含量明显升高,以及胃窦粘膜中GAS表达显著上调,下丘脑和血清中脑肠肽CCK-8、SS含量明显降低,以及胃窦粘膜中SS表达均显著下调,胃粘膜充血情况修复,胃、十二指肠表层上皮细胞变性坏死及松解脱落情况改善。结论:四磨汤对幼龄厌食症小鼠有明显治疗作用,作用机制可能与其降低脑肠肽CCK-8、SS,促进MTL、GAS等胃肠激素的释放和胃肠粘膜生长,加强胃肠动力有关。 Objective:To study the therapeutic effect of Simo Tang on anorexia model in young mice and its possible mechanism.Methods:Anorexia model in young mice was established by special feed.Effects on the body weight,the food intake and the gastrointestinal motility were observed in different doses of Simo Tang(5 mL/kg、10 mL/kg、20 mL/kg)groups.HE staining was used to observe the pathological changes of gastrointestinal tract.Levels of CCK-8,SS,β-EP in the hypothalamus and serum,and GAS and MTL in the serum were detected by ELISA.Expression levels of SS cells and GAS cells in gastric antral mucosa were detected by immunohistochemistry.Results:Compared with the control group,the body weight growth,the food intake,the gastrointestinal motility,the levels of serum gastrointestinal hormones MTL,GAS,and GAS expression in gastric antral mucosa were significantly reduced in the model group.Contents of CCK-8 and SS in hypothalamus and serum were increased,the expression of SS in gastric antral mucosa was up regulated in the model group.Also,the hyperemic gastric mucosa,the degeneration and necrosis of the epithelial cells in stomach and duodenum were observed.Compared with the model group,after administered with 20 mL/kg Simo Tang for 2 weeks,the body weight growth,the food intake,the gastrointestinal motility,the levels of serum gastrointestinal hormones MTL,GAS,and GAS expression in gastric antral mucosa were increased,the contents of CCK-8 and SS in hypothalamus and serum were significantly decreased,the expression of SS in the mucosa was significantly down regulated.The congestive condition of the gastric mucosa was repaired,and the degeneration and necrosis of the epithelial cells in stomach and duodenum was improved.Conclusion:Simo Tang has the obvious therapeutic effect on anorexia model in young mice.The mechanism may be related to lowering brain-gut peptide CCK-8 and SS,promoting the release of gastrointestinal hormones such as MTL and GAS,promoting the growth of gastrointestinal mucosa and strengthening gastrointestinal motility.
