摘要
目的探讨ULA-G5-URR基因多态性在子痫前期患者中的表达及与血浆可溶性HLA-G(sHLA-G)水平的相关性。方法选择2016年5月-2018年12月治疗的子痫前期患者67例作为对象,设为观察组;选择同期正常检查孕妇58例,设为对照组。两组入院后均完成外周静脉血5mL采集,利用全血基因DNA、聚合酶链式反应技术(PCR)、琼脂糖电泳及基因位点测序等方法完成两组ULA-G5-URR基因多态性测定;采用酶联免疫吸附试验完成两组sHLA-G水平测定;采用SPSS Pearson相关性分析软件对子痫前期患者ULA-G5-URR基因位点与sHLA-G水平进行相关性分析。结果观察组子痫前期患者ULA-G5-URR基因SPN位点中+274位点与+289位点率,均高于对照组(P<0.05);观察组子痫前期+274位点与+29位点率中A位点,均高于对照组(P<0.05);观察组子痫前期+274位点与+289位点率中G/T位点低于对照组(P<0.05);观察组sHLA-G水平高于对照组(P<0.05);观察组中重度子痫前期sHLA-G水平,高于轻度子痫与中度子痫(P<0.05);观察组中中度子痫前期sHLA-G水平,高于轻度子痫(P<0.05);SPSS Pearson相关性分析结果表明:子痫前期患者sHLA-G水平与+274位点与+289位点呈正相关性(P<0.05)。结论ULA-G5-URR基因中+274位点与+289位点多态性与子痫前期发生、发展有关;sHLA-G水平在子痫前期患者中呈高表达,其表达水平与ULA-G5-URR基因多态性存在相关性。
Objective:To investigate the expression of ULA-G5-URR gene polymorphism in preeclamptic patients and its correlation with plasma soluble HLA-G(sHLA-G)levels.Methods:Sixty-seven patients with preeclampsia treated from May 2016 to December 2018 were selected as the observation group.58 patients with normal pregnancy were selected as the control group.Two groups of peripheral venous blood were collected from the two groups after admission.The two groups of ULA-G5-URR gene polymorphisms were completed by whole blood gene DNA,polymerase chain reaction(PCR),agarose electrophoresis and gene locus sequencing.The sHLA-G levels were determined by enzyme-linked immunosorbent assay(ELISA).The correlation between ULA-G5-URR loci and sHLA-G levels in preeclampsia patients was analyzed by SPSS Pearson correlation analysis software.Results:The rate of+274 and+289 in the SPN locus of ULA-G5-URR gene in the preeclampsia group was higher than that in the control group(P<0.05).The preeclampsia+274 locus in the observation group The A site in the+29 locus rate was higher than the control group(P<0.05).The G/T locus in the preeclampsia+274 and+289 loci was lower in the observation group than in the control group(P<0.05).The level of sHLA-G in the observation group was higher than that in the control group(P<0.05);the level of sHLA-G in the pre-eclampsia was higher in the observation group than in mild eclampsia and moderate eclampsia(P<0.05);The level of sHLA-G in moderate preeclampsia was higher than that in mild eclampsia(P<0.05).The results of SPSS Pearson correlation analysis showed that the level of sHLA-G in patients with preeclampsia was positively correlated with+274 and+289.Sex(P<0.05).Conclusion:The+274 and+289 loci polymorphisms in ULA-G5-URR gene are associated with the development and progression of preeclampsia;sHLA-G levels are highly expressed in preeclampsia patients,and their expression levels are consistent with ULA-There is a correlation between G5-URR gene polymorphisms.
