润肠通秘茶的超高效液相色谱指纹图谱研究及其6种成分含量测定

摘要 目的建立润肠通秘茶的超高效液相色谱(UPLC)指纹图谱,并对其6种成分进行含量测定。方法以UPLC技术建立润肠通秘茶的指纹图谱及测定其6种成分的含量。采用Ultimate UHPLC AQ-C_(18)色谱柱(2.1 mm×100 mm,1.8μm),以乙腈-0.1%甲酸水为流动相,梯度洗脱;流速:0.5 mL·min^(-1);柱温:28℃;检测波长:280 nm。采用《中药色谱指纹图谱相似度评价系统》(2012年版)、SPSS 25统计软件及SIMCA14.1软件对数据进行相似度分析、聚类分析(CA)及主成分分析(PCA)。结果建立了润肠通秘茶的UPLC指纹图谱,确定了14个共有峰,且相似度在1~0.992范围内。含量测定结果显示,松果菊苷(5号峰)、阿魏酸(6号峰)、毛蕊异黄酮葡萄糖苷(7号峰)、毛蕊花糖苷(8号峰)、橙皮苷(12号峰)、芒柄花素(14号峰)分别在272.500~4.258、67.500~1.055、76.250~1.191、300.000~4.688、292.500~4.570、30.625~0.479μg·mL^(-1)范围内呈良好的线性关系;平均回收率分别为102.47%、103.98%、94.42%、97.00%、103.62%、96.09%;11批润肠通秘茶中上述各成分的质量分数分别为:5.8553~6.1972、0.0861~0.0959、0.1374~0.1774、0.3716~0.4385、4.9258~6.8984、0.0480~0.0511 mg·g^(-1)。聚类分析和主成分分析均显示11批润肠通秘茶可以分为3类。结论所建立的润肠通秘茶UPLC指纹图谱及化学成分含量测定方法可以简单、快速地对不同批次的润肠通秘茶进行质量检测,为其质量控制提供参考。 Objective To establish the ultra-high performance liquid chromatography(UPLC)fingerprint of Runchang Tongmi Tea and to determine the contents of six components.Methods UPLC technology was used to establish the fingerprint of Runchang Tongmi Tea and determine the content of its six components.Analysis was carried out on an Ultimate UHPLC AQ-C_(18)column(2.1 mm×100 mm,1.8μm)using acetonitrile-0.1%formic acid(aq)as the mobile phase for gradient elution.Flow velocity:0.5 mL·min^(-1),Column temperature:28℃,and detection wavelength:280 nm.Similarity Evaluation System for Chromatographic Fingerprint of TCM(version 2012),SPSS 25 statistical software and SIMCA 14.1 software were used for similarity analysis,cluster analysis(CA)and principal component analysis(PCA).Results The UPLC fingerprint of Runchang Tongmi Tea was established.The 14 common peaks were identified,and the similarity was in the range of 1-0.992.The content analysis demonstrated that echinacoside(peak No.5),ferulic acid(peak No.6),calycosin-7-O-β-D-glucoside(peak No.7),acteoside(peak No.8),hesperidin(peak No.12)and formononetin(peak No.14)showed good linearity in the ranges of 272.500-4.258,67.500-1.055,76.250-1.191,300.000-4.688,292.500-4.570,and 30.625-0.479μg·mL^(-1),respectively.The average recovery rates were 102.47%,103.98%,94.42%,97.00%,103.62%and 96.09%.The contents of above mentioned components in 11 batches of Runchang Tongmi Tea were 5.8553-6.1972,0.0861-0.0959,0.1374-0.1774,0.3716-0.4385,4.9258-6.8984 and 0.0480-0.0511 mg·g^(-1).Data from CA and PCA showed that 11 batches of RunChang Tongmi Tea can be divided into 3 categories.Conclusion The establishment of Runchang Tongmi Tea UPLC fingerprint map and chemical composition content measurement method can be simply and rapidly detecting the quality of Runchang Tongmi Tea of different batches to provide a reference for its quality control.
