Preliminary Exploration of Antidepressant Mechanism of Catalpol from Dihuang(Rehmannia glutinosa)

摘要 目的从抗氧化防御探索地黄梓醇的抗抑郁机制。方法在前期证实梓醇具有抗抑郁药效的基础上,通过对慢性不可预知温和应激(CUMS)抑郁症大鼠额叶皮层的部分氧化和抗氧化指标的检测分析,评价梓醇抗抑郁的机制。结果与正常组比较,CUMS引起了大鼠额叶皮层的脂质过氧化产物丙二醛(MDA)水平显著升高,主要抗氧化酶超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、抗氧化物质还原型谷胱甘肽(GSH)、谷胱甘肽相关抗氧化酶谷胱甘肽过氧化物酶(GPx)和谷胱甘肽-S转移酶(GST)的水平均显著降低;而给予梓醇5周后,CUMS引起的以上指标的异常均被显著逆转。结论梓醇的抗抑郁机制可能与抑制额叶皮层脂质过氧化并增强其抗氧化水平,进而纠正氧化/抗氧化失衡有关。 Objective To initially explore the antidepressant mechanism of catalpol from Dihuang(Rehmannia glutinosa)based on the antioxidant defense.Methods It evaluated the antidepressant mechanism of catalpol by detecting and analyzing the partial oxidation and antioxidant indexes of the frontal cortex in rats undergoing chronic unpredictable mild stress(CUMS)on the basis of our previous confirmation that catalpol had antidepressant effects.Results Compared with the normal group,CUMS caused a significant increase in the level of the lipid peroxidation product malondialdehyde(MDA)in the frontal cortex of rats.The levels of the major antioxidant enzymes superoxide dismutase(SOD)and catalase(CAT)and the levels of antioxidant substance reduced glutathione(GSH),glutathione-related antioxidant enzyme glutathione peroxidase(GPx)and glutathione-S transferase(GST)were significantly decreased.Fortunately,after catalpol was administered to the rats for 5 weeks,the abnormalities of the above indicators caused by CUMS were significantly reversed.Conclusion The antidepressant mechanism of catalpol may be related to inhibiting the lipid peroxidation of the frontal cortex and enhancing its antioxidant level,thereby correcting the oxidation/antioxidation imbalance.
