凉粉草不同种质的染色体倍性与基因组大小研究

摘要 目的:建立凉粉草基于流式细胞术的染色体倍性鉴定技术与基因组大小检测方法。方法:以收集的36份凉粉草栽培种质及1份野生种质为材料,采用流式细胞术估测凉粉草基因组大小,并以二倍体为对照,计算凉粉草染色体倍性。结果:内参与待测样品峰值能完全分开,无重叠峰,峰形清晰集中,可对凉粉草基因组大小进行有效估测。鉴定凉粉草供试材料中12份为二倍体,基因组大小为1.14~1.35 Gb;20份为三倍体,基因组大小为1.74~2.01 Gb;5份为四倍体,基因组大小为2.42~2.47 Gb。结论:成功建立了凉粉草基于流式细胞术的染色体倍性鉴定技术与基因组大小检测方法,凉粉草不同种质倍性水平丰富,不同凉粉草种质的倍性水平与其性状具有显著相关性。 Objective:To establish chromosomal ploidy identification technology and genome size detection method of Mesona chinensis based on flow cytometry.Methods:The genome size of thirty-six cultivated germplasm and one wild germplasm was estimated by flow cytometry,and the ploidy was calculated using diploid as the control.Results:The peak values of the internal parameter and need cheeking samples could be separated completely without overlapping peaks,and the peak shape was clear and concentrated,which could effectively estimate the genome size.Twelve germplasms were diploid with genome sizes between 1.14 to 1.35 Gb.Twenty germplasms were triploid with the genome size between 1.74 to 2.01 Gb.Five germplasms were tetraploids with genome sizes between 2.42 to 2.47 Gb.Conclusion:The chromosomal ploidy identification technigne and gerome size detection method of Mesona chinensis are established successfully based on flow-cytometry.The ploidy levels of different germplasm of Mesona chinensis are abundant,and the ploidy levels of different species of Mesona chinensis are significantly correlated with the traits.
