Analysis of Inorganic Elements in Eucommiae Folium by ICP-MS Combined with Chemometrics and Health Risk Assessment

摘要 目的:测定杜仲叶中18个无机元素的含量,研究杜仲叶生长过程中无机元素变化规律以及不同栽培模式、不同产地杜仲叶无机元素含量差异,并对其5个重金属及有害元素进行健康风险评估。方法:采用微波消解法对杜仲叶样品进行处理,通过ICP-MS法测定As、Cd、Cr、Cu、Mn、Zn、Al、Pb、Fe等18个无机元素的含量,分析不同采收期杜仲叶无机元素变化规律,利用正交偏最小二乘判别分析(OPLS-DA)比较不同产地、不同栽培模式杜仲叶无机元素差异,并通过计算重金属元素每日最大可耐受量(EDI)、靶标危害系数(THQ)和致癌风险(CR)进行健康风险评估。结果:杜仲叶无机元素含量与采收期、栽培模式和产地有一定联系,不同栽培模式和产地杜仲叶无机元素含量差异显著,且重金属健康风险评估模型EDI、THQ、CR结果显示27批杜仲叶未对暴露人群造成明显的健康危害,不存在明显的致癌风险。结论:杜仲叶无机元素种类含量丰富,其重金属不存在明显的健康风险,该结果可为杜仲叶的食药两用安全性研究提供参考。 Objective:To determine the contents of 18 inorganic elements contents in Eucommiae Folium,to study the changes of inorganic elements in Eucommiae Folium during the growth process and the differences of inorganic elements contents in Eucommiae Folium from different cultivation modes and different habitats,and to assess the health risks of 5 heavy metals and harmful elements.Methods:The samples of Eucommiae Folium were processed by microwave digestion,and the contents of 18 inorganic elements,such as As,Cd,Cr,Cu,Mn,Zn,Al,Pb,Fe were determined by ICP-MS.The changes of inorganic elements in Eucommiae Folium in different harvesting periods were analyzed.Orthogonal partial least squares discriminant analysis(OPLS-DA)was used to compare the differences of inorganic elements in Eucommiae Folium from different habitats and cultivation modes,and the health risk was assessed by calculating the maximum tolerable daily amount(EDI),target hazard factor(THQ)and carcinogenic risk(CR)of heavy metals.Results:The contents of inorganic elements in Eucommiae Folium were related to harvesting period,cultivation mode and producing area.The inorganic elements contents of Eucommiae Folium varied significantly in different cultivation modes and producing areas.The results of EDI,THQ and CR of heavy metal health risk assessment models showed that the 27 batches of Eucommiae Folium did not cause significant health harm to the exposed population,and there was no significant carcinogenic risk.Conclusion:Eucommiae Folium is rich in inorganic elements and has no obvious health risk of heavy metals,the results can provide reference for the safety of Eucommiae Folium for both food and medicine.
