硫熏对粉葛中无机元素与异黄酮类成分含量的影响 被引量:1

Effect of Sulfur Fumigation on the Contents of Inorganic Elements and Isoflavones in Puerariae Thomsonii Radix

摘要 目的:探究硫熏前后粉葛中无机元素与异黄酮类成分含量变化的规律。方法:采用酸碱滴定法测定硫熏粉葛中二氧化硫残留量;ICP-MS法测定硫熏前后粉葛中Cr、Mo、Pb、Cd、V、Mn、Cu、Sr等22个无机元素含量;HPLC法测定硫熏前后粉葛中4′-O-甲基葛根素、大豆苷、黄豆黄苷、大豆苷元、葛根素等5个异黄酮类成分的含量;对硫熏前后发生显著变化的无机元素与异黄酮类成分进行冗余分析。结果:10批硫熏粉葛样品二氧化硫残留量为1281.01~2570.48 mg/kg,均超过限度。粉葛中Cr、Mo、Cd、Pb等元素在硫熏后含量极显著增加(P<0.01),而V、Mn、Cu、Sr等含量显著或极显著降低(P<0.05或P<0.01)。粉葛经硫熏后,4′-O-甲基葛根素、大豆苷、黄豆黄苷和大豆苷元等含量显著或极显著降低(P<0.05或P<0.01),而葛根素含量显著增加(P<0.05)。冗余分析结果表明Cr、Pb、Cd等3个元素对粉葛中异黄酮类成分含量变化影响较大(P<0.05或P<0.01)。结论:硫熏可改变粉葛中无机元素和异黄酮类成分的含量,结合冗余分析可真实反映硫熏药材的本质,可为评价粉葛的质量提供依据。 Objective:To explore the changes of inorganic elements and isoflavones in Puerariae Thomsonii Radix before and after sulfur-fumigation.Methods:The sulfur dioxide residues in sulfur-fumigated Puerariae Thomsonii Radix were determined by acid-base titration.ICP-MS method was used to determine the contents of twenty-two inorganic elements including Cr,Mo,Pb,Cd,V,Mn,Cu,Sr in Puerariae Thomsonii Radix before and after sulfur fumigation.HPLC method was used to determine the contents of five isoflavones including 4′-O-methyl puerarin,daidzin,glycitin,daidzein,puerarin in Puerariae Thomsonii Radix before and after sulfur fumigation.Redundancy analysis of inorganic elements and isoflavones which changed significantly in Puerariae Thomsonii Radix before and after sulfur fumigation was carried out.Results:The residual amount of sulfur dioxide in 10 batches of sulfur-fumigated Puerariae Thomsonii Radix ranged from 1281.01 to 2570.48 mg/kg,all exceeding the limit.After sulfur fumigation,the contents of Cr,Mo,Cd and Pb were significantly increased(P<0.01),while the contents of V,Mn,Cu and Sr were significantly decreased(P<0.05 or P<0.01),the contents of 4′-O-methyl puerarin,daidzin,glycitin and daidzein significcantly decreased(P<0.05 or P<0.01),while the content of puerarin significcantly increased(P<0.05).The results of redundancy analysis showed that Cr,Pb and Cd had a significant effect on isoflavones content in Puerariae Thomsonii Radix(P<0.05 or P<0.01).Conclusion:Sulfur fumigation can change the content of inorganic elements and isoflavones in Puerariae Thomsonii Radix.Combined with redundancy analysis,the essence of sulfur fumigation can be truly reflected,which can provide a basis for the evaluation of the quality of Puerariae Thomsonii Radix.
