柚皮素通过调控L1细胞黏附分子表达对Ⅱ型肺泡上皮细胞间质转分化的影响

摘要 目的:观察柚皮素对Ⅱ型肺泡上皮细胞间质转分化的影响,并探讨其作用机制。方法:采用CCK-8法检测不同浓度柚皮素(0、12.5、25、50、100、200、400μmol/L)对Ⅱ型肺泡上皮细胞增殖活性的影响。采用5 ng/mL TGF-β_(1)刺激Ⅱ型肺泡上皮细胞建立上皮间质转分化模型,并加入不同浓度柚皮素(12.5、25、50μmol/L)进行干预。倒置显微镜下观察细胞形态变化;qRT-PCR和Western Blot法检测细胞中L1细胞黏附分子(L1CAM)、上皮间质转分化相关基因E-cadherin、Vimentin、N-cadherin及纤维化相关基因collagenⅠ、α-SMA mRNA和蛋白表达;构建L1CAM过表达的Ⅱ型肺泡上皮细胞,再采用5 ng/mL TGF-β_(1)或联合50μmol/L柚皮素进行干预,qRT-PCR和Western Blot检测细胞中上述相关基因mRNA及蛋白表达。结果:当柚皮素浓度>50μmol/L时其对Ⅱ型肺泡上皮细胞增殖活性有显著抑制作用。与空白对照组比较,模型组Ⅱ型肺泡上皮细胞呈梭形或纺锤形变化,细胞中E-cadherin mRNA及蛋白表达显著降低,L1CAM、Vimentin、N-cadherin、collagenⅠ、α-SMA mRNA及蛋白表达显著升高(P<0.05)。与模型组比较,柚皮素组Ⅱ型肺泡上皮细胞形态未发生明显变化,细胞中E-cadherin mRNA和蛋白表达显著升高,L1CAM、Vimentin、N-cadherin、collagenⅠ、α-SMA mRNA及蛋白表达显著降低(P<0.05)。L1CAM过表达可显著逆转柚皮素对Ⅱ型肺泡上皮细胞间质转分化的抑制作用。结论:柚皮素可抑制TGF-β_(1)诱导的Ⅱ型肺泡上皮细胞间质转分化,其作用机制可能与下调L1CAM表达有关。
