基于UPLC-Q-TOF-MS/MS的桑白皮大鼠体内入血成分研究 被引量:7

摘要 目的:采用超高效液相色谱-四极杆-飞行时间质谱联用技术(UPLC-Q-TOF-MS/MS)初步分析大鼠口服给药桑白皮后的入血成分。方法:采用ACQUITY UPLC BEH C_(18)(100 mm×2.1 mm,1.7μm)色谱柱;流动相为0.1%甲酸水溶液-0.1%甲酸乙腈溶液,梯度洗脱;流速400μL/min;柱温40℃;进样量5μL。采用电喷雾离子源,正、负离子模式下采集一、二级质谱数据,利用自建二级质谱数据库及相应裂解规律匹配法,结合文献数据鉴定血清中化学成分。结果:在给药后的大鼠血清中分析表征出了10个化学成分,均为原型成分,依次为:3,4-二羟基苯甲醛、咖啡酸、2,4-二羟基苯甲酸、秦皮乙素、葛根素、7-羟基香豆素、壬二酸、桑色素、桑辛素P、6-姜酚。结论:血清中鉴定出的原型成分可能是桑白皮潜在的活性成分,为其药效物质基础研究提供了依据。 Objective:To preliminarily analyze the chemical components in rat serum after oral administration of Mori Cortex by UPLC-Q-TOF-MS/MS.Methods:The ACQUITY UPLC BEH C_(18)(100 mm×2.1 mm,1.7μm)column was used for isolation and identification of chemical components in Mori Cortex with a mobile phase of 0.1%formic acid solution and acetonitrile containing 0.1%formic acid for gradient elution and the flow rate was 400μL/min.The column temperature was 40℃and injection volume was 5μL.The primary and secondary mass spectrometry data were collected in positive and negative ion mode using electrospray ionization(ESI)ion source.The chemical constituents in serum were identified by using the self-built secondary mass spectrometry database and the corresponding cracking law matching method combined with literature data.Results:Ten chemical constituents were identified in rat serum after administration,all of which were prototype components,including 3,4-dihydroxybenzaldehyde,caffeic acid,2,4-dihydroxybenzoic acid,aesculetin,puerarin,7-hydroxycoumarin,azelaic acid,morin,morin P,6-gingerol.Conclusion:The prototype components identified in serum may be the potential active components of Mori Cortex,which provide a basis for the basic research of its pharmacodynamic substances.
