基于指纹图谱的黄芪“临床重用”与“小剂量久煎”一致性评价研究 被引量:1

Study on Consistency Evaluation of "Clinical Reuse" and "Low-dose Long-decoction" of Astragali Radix Based on Fingerprint

摘要 目的:建立临床重用90 g与小剂量30 g久煎的黄芪水煎液指纹图谱,并进行一致性评价研究。方法:采用HPLC-ELSD法建立黄芪90 g提取50 min和30 g提取6个不同时间(50、100、150、200、250、300 min)的水煎液指纹图谱;利用聚类分析(HCA)和偏最小二乘判别分析(PLS-DA)对共有峰峰面积进行分析,并进行黄芪甲苷的含量测定,研究化学成分变化情况。结果:70批黄芪水煎液HPLC-ELSD指纹图谱中有6个共有峰,指认了其中3个成分,分别为黄芪甲苷、黄芪皂苷Ⅱ、黄芪皂苷Ⅰ,HPLC-ELSD指纹图谱相似度为0.758~0.978。聚类分析与偏最小二乘判别分析结果一致,黄芪重用90 g提取50 min和黄芪30 g提取300 min聚为一类。结论:黄芪小剂量30 g提取300 min时可与重用90 g提取50 min水煎液指纹图谱较一致,临床使用中单味黄芪可适当延长提取时间,复方中黄芪或可先煎、久煎代替重用。 Objective:To establish the fingerprints of the decoction of Astragali Radix with clinical reuse of 90 g and a small dose of 30 g for a long time,and to evaluate the consistency.Methods:HPLC-ELSD method was used to establish the fingerprints of the decoction of Astragali Radix extracted at 90 g for 50 min and 30 g for 6 different extraction times,including 50,100,150,200,250,300 min.Cluster analysis(HCA)and partial least square discriminant analysis(PLS-DA)were used to analyze the common peak areas,and the content of astragalosideⅣwas determined to study the chemical composition changes.Results:There were 6 common peaks in the HPLC-ELSD fingerprints of the 70 batches of Astragali Radix water decoction,and 3 of them were identified as astragalosideⅣ,astragalosideⅡand astragalosideⅠ,respectively.The similarity of HPLC-ELSD fingerprints ranged from 0.758 to 0.978.The results of cluster analysis and partial least squares discriminant analysis were consistent.The Astragali Radix which was reused for 90 g extraction for 50 min and Astragali Radix which was extracted for 30 g extraction for 300 min were clustered into one class.Conclusion:The fingerprint of Astragali Radix extracted at a small dose of 30 g for 300 min is consistent with the fingerprint of water decoction extracted at 90 g for 50 min.In clinical use,the extraction time of single Astragali Radix can be appropriately prolonged,and the Astragali Radix in compound may be replaced by decocting first or long decocting.
