加速实验过程牛膝成分含量与颜色值相关性研究 被引量:1

摘要 目的:研究牛膝加速实验过程中成分含量变化及其与颜色值的相关性。方法:为期6个月加速实验获取牛膝药材及饮片样品,通过视觉分析仪(电子眼)量化牛膝药材表面及饮片粉末颜色值,采用HPLC法测定β-蜕皮甾酮、25R-牛膝甾酮、25S-牛膝甾酮含量,按照中国药典方法测定醇溶性浸出物含量,苯酚硫酸法测定多糖含量,应用相关性分析、回归分析等方法分析颜色值与成分含量数据。结果:牛膝药材表面及饮片粉末L^(*)值随加速实验延长整体呈降低趋势。L^(*)、a^(*)、b^(*)值与药材浸出物含量存在极显著负相关,与多糖含量存在极显著正相关(P<0.01);与饮片β-蜕皮甾酮、25R-牛膝甾酮、25S-牛膝甾酮、多糖含量呈显著相关(P<0.05)。回归分析结果得出药材浸出物和多糖预测回归曲线。结论:L^(*)可作为牛膝贮藏期间质量控制监测指标,为建立牛膝颜色从经验转向客观量化的评价模式奠定基础。 Objective:To study the changes of component contents and its correlation with color values of Achyranthis Bidentatae Radix during the accelerated test.Methods:The samples of Achyranthis Bidentatae Radix medicinal material and decoction pieces were obtained by accelerated test for 6 months,and the color values of the Achyranthis Bidentatae Radix medicinal material surface and decoction pieces powder were quantified through the electronic-eye,HPLC method was used to determine the contents ofβ-ecdone,25R-inokosterone and 25S-inokosterone.The content of the extract was determined by alcohol-soluble extract method in pharmacopeia,and the content of polysaccharide was determined by phenol sulfuric acid method.Correlation analysis,regression analysis and other methods were used to analyze the color values and component contents data.Results:The L^(*)values of Achyranthis Bidentatae Radix medicinal material surface and decoction pieces powder decreased with the extension of accelerated test.The L^(*),a^(*),b^(*)values were significantly negatively correlated with extract content of medicinal material,significantly positively correlated with polysaccharide content of medicinal materials(P<0.01)and significantly correlated with the contents ofβ-ecdone,25R-inokosterone,25S-inokosterone and polysaccharide of decoction pieces(P<0.05).The regression curves of extracts and polysaccharides were obtained by regression analysis.Conclusion:The L^(*)value can be used as a monitoring index for quality control of Achyranthis Bidentatae Radix during storage,laying a foundation for the evaluation model of color of Achyranthis Bidentatae Radix from experience to objective quantification.
