摘要
目的:研究环境因子对栀子西红花苷积累及其合成相关基因表达的影响。方法:以江西信丰、永丰、樟树、丰城、共青城5个地区栀子果实为材料,使用HPLC测定不同地区栀子果实中的西红花苷-Ⅰ含量,并用荧光定量PCR测定西红花苷相关基因表达量(PSY、CCD4、NCED6、GGPS2、UGT86A1、UGT85A2);利用Pearson系数结合环境因子进行关联分析。结果:土壤中的有效硼、速效磷以及相对湿度与栀子果实中西红花苷-Ⅰ含量具有极显著相关性(P<0.01),栀子PSY与UGT85A2基因的表达量与大叶大果型栀子西红花苷-Ⅰ含量呈极显著正相关(P<0.01)。结论:土壤中的有效硼、速效磷以及相对湿度是影响栀子果实中西红花苷-Ⅰ合成的主导环境因子。
Objective:To study the effects of environmental factors on the accumulation of crocin and expression of genes related to crocin synthesis in Gardenia jasminoides.Methods:Using Gardenia jasminoides fruits from Xinfeng,Yongfeng,Zhangshu,Fengcheng and Gongqing city in Jiangxi as materials,the content of crocin-Ⅰin Gardenia jasminoides fruits from different regions was determined by HPLC,and the expressions of crocin related genes including PSY,CCD4,NCED6,GGPS2,UGT86A1,UGT85A2 were determined by fluorescent quantitative PCR.Pearson coefficient and environmental factors were used for correlation analysis.Results:There was a significant correlation between available boron,available phosphorus,relative humidity and content of crocin-Ⅰin Gardenia jasminoides fruits(P<0.01).The expressions of PSY and UGT85A2 genes were positively correlated with the content of crocin-Ⅰin Gardenia jasminoides with large leaves and fruits(P<0.01).Conclusion:Available boron,available phosphorus and relative humidity in soil are the main environmental factors affecting crocin-Ⅰsynthesis in Gardenia jasminoides fruits.
