单叶蔓荆果实的化学成分研究

Chemical Constituents from the Fruits of Vitex trifolia var. simplicifolia

目的:对马鞭草科牡荆属植物单叶蔓荆Vitex trifolia L.var.simplicifolia Cham(Vitex rotundifolia L.)的成熟果实蔓荆子乙醇提取物的正丁醇部位化学成分进行研究。方法:通过硅胶柱层析、ODS柱色谱、Sephadex LH-20凝胶柱色谱和制备液相等色谱技术进行分离纯化,并应用核磁共振(NMR)以及质谱等波谱技术进行化合物结构鉴定。结果:从单叶蔓荆果实乙醇提取物的正丁醇部位分离得到12个化合物,分别鉴定为:5,6,7,8,4′-pentamethoxyflavone(1)、牡荆素(2)、2″-O-鼠李糖牡荆素(3)、5-去甲基甜橙黄酮(4)、开环异落叶松树脂酚(5)、(7R,8S)-dihydrodehydrodiconiferyl alcohol-9′-O-β-D-glucopyranoside(6)、3-(β-D-glucopyranosyloxymethyl)-2-(4-hydroxy-3-methoxyphenyl)-5-(3-hydroxypropyl)-7-methoxydihydrobenzofuran(7)、异嗪皮啶(8)、5-methoxy-6,7-methylene-dioxy coumarin(9)、(3S,5S,8R,9R,10S)-3,9-dihydroxy-13(14)-labdaen-16,15-olide 3-O-β-D-glucopyranoside(10)、vitexilactone(11)、pedunculariside(12)。结论:除化合物2、11、12外,其余化合物均为首次从该植物中分离得到。

Objective:To study the chemical constituents from the n-butanol fraction of ethanol extract of Vitex trifolia var.simplicifolia fruits.Methods:The compounds were isolated and purified by silica gel column chromatography,ODS column chromatography,Sephadex LH-20 column chromatography and preparation liquid chromatography.The structures of the compounds were identified by NMR and mass spectrometry.Results:Twelve compounds were isolated from the n-butanol fraction of ethanol extract of Vitex trifolia var.simplicifolia fruits and identified as 5,6,7,8,4′-pentamethoxyflavone(1),vitexin(2),2″-O-rhamnosyl vitexin(3),5-hydroxy-3′,4′,6,9-tetramethoxy flavonol(4),secoisolariciresinol(5),(7R,8S)-dihydrodehydrodiconiferyl alcohol-9′-O-β-D-glucopyranoside(6),3-(β-D-glucopyranosyloxymethyl)-2-(4-hydroxy-3-methoxyphenyl)-5-(3-hydroxypropyl)-7-methoxydihydrobenzofuran(7),isofraxidin(8),5-methoxy-6,7-methylene-dioxy coumarin(9),(3S,5S,8R,9R,10S)-3,9-dihydroxy-13(14)-labdaen-16,15-olide 3-O-β-D-glucopyranoside(10),vitexilactone(11),pedunculariside(12),respectively.Conclusion:Except for compounds 2,11,12,other compounds are isolated from Vitex trifolia var.simplicifolia for the first time.
