基于HS-SPME-GC-MS分析不同产地初加工方式对山银花挥发性成分的影响

The Analysis of the Effects of Different Primary Processing Methods on Volatile Components of Lonicerae Flos via HS-SPME-GC-MS

目的:分析山银花中的挥发性成分并评价不同初加工方式对挥发性成分的影响。方法:基于顶空固相微萃取串联气相色谱-质谱联用(HS-SPME-GC-MS)技术鉴定“湘蕾”和“金翠蕾”山银花中的挥发性成分,并采用主成分分析法(PCA)评价不同初加工方式对其挥发性成分的影响。结果:“湘蕾”和“金翠蕾”山银花分别含有33个和20个匹配度>85%的挥发性成分,主要由烯醇、直链烷烃、醛、酯等4类化合物构成,且以苯乙醇、1-辛醇、2H-pyran-3-ol,6-ethenyltetrahydro-2,2,6-trimethyl-,(3R,6S)-rel-和1-癸醇为主。2种山银花经过直接冷冻干燥、45℃直接干燥、蒸气杀青(45℃干燥)、滚筒杀青(45℃干燥)等4种处理后,通过PCA分析得出冷冻干燥对“湘蕾”中挥发性成分的影响不显著,45℃直接干燥对“金翠蕾”中挥发性成分的影响不显著。结论:该研究可为山银花中化学成分的研究及其进一步开发提供参考。

Objective:To analyze the volatile components in Lonicerae Flos and evaluate the effects of different primary processing methods on the volatile components.Methods:The headspace solid phase microextraction coupled with gas chromatography-mass spectrometry(HS-SPME-GC-MS)was used to analyze the volatile components in two varieties of Lonicerae Flos“Xianglei”and“Jincuilei”,principal component analysis(PCA)was applied to evaluate the effects of different primary processing methods on the volatile components.Results:There were 33 and 20 kinds of volatile components with the match of more than 85%in Lonicerae Flos"Xianglei"and"Jincuilei",respectively,which were mainly composed of enols,straight chain alkanes,aldehydes and esters.The main components were phenethyl alcohol,1-octanol,2 H-pyran-3-ol,6-ethenyltetrahydro-2,2,6-trimethyl-,(3R,6S)-rel-and decyl alcohol.The two kinds of Lonicerae Flos were treated by direct freeze-drying,45℃direct drying,inactivating the enzyme by steam(drying at 45℃)and by roller(drying at 45℃).PCA indicated that the effect of freeze-drying on the volatile components of"Xianglei"was not significant,and the effect of direct drying at 45℃on the volatile components of"Jincuilei"was not significant.Conclusion:This study can provide reference for the study and further development of chemical constituents in Lonicerae Flos.
